杂豆膳食纤维对糖尿病大鼠胰腺氧化损伤的修复作用 被引量:3

Effect of miscellaneous bean dietary fiber on pancreatic oxidative damage repair of diabetic rats

摘要 目的探讨不同剂量杂豆膳食纤维对糖尿病模型大鼠胰腺氧化损伤的修复作用。方法经腹腔注射链脲佐菌素(streptozocin,STZ)溶液(150 mg/kg)构建糖尿病大鼠模型,建模成功后,连续灌胃低、中、高剂量(1.35、2.7、5.4 g/kg)杂豆膳食纤维42 d,并设对照组(灌胃格列本脲)、空白组(灌胃生理盐水)及模型组(灌胃生理盐水)。测定各组大鼠体重、胰腺体比、血清中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)及丙二醛(malonic dialdehyde,MDA)含量,并取胰腺组织制作H&E染色病理学切片进行观察。结果与模型组大鼠相比,各灌胃剂量组大鼠体重均升高,胰腺体比均不同程度下降,血清GSH-Px及SOD含量均不同程度上升,血清MDA含量均不同程度下降,且胰腺组织损伤程度明显减轻,胰岛细胞数目增多,细胞水肿变形程度减轻。结论杂豆膳食纤维可修复糖尿病对大鼠的氧化损伤,改善对大鼠胰腺的损伤情况,且改善效果与剂量呈正相关。 Objective To investigate the effect of miscellaneous bean dietary fiber at various dosages on pancreatic oxidative damage repair of diabetic rats. Methods Diabetic rat model was established by intraperitoneal injection with streptozocin(STZ) at a dosage of 150 mg/kg. The model rats were treated with miscellaneous bean dietary fiber at low(1. 35 g/kg),moderate(2. 7 g/kg)and high(5. 4 g/kg)dosages by gavage for 42 d. The healthy rats as normal control and the model rats as model control were treated with physiological saline,while the model rats in parallel control group with glyburide. The bodyweight,pancreatic viscera ratio as well as superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malonic dialdehyde(MDA)contents in sera in various groups were measured,while the pancreatic tissues were prepared into pathological sections and observed by HE staining. Results Compared with those in model control group,the body weight of rats treated with miscellaneous bean dietary fiber at various dosages increased,while the pancreatic viscera ratio decreased,the serum GSH-Px and SOD contents increased,the serum MDA content decreased,and the damage of pancreatic tissue was relieved. The islet cells increased in quantity,of which the swelling and deformation were relieved. Conclusion Miscellaneous bean dietary fiber may repair the oxidative damage in diabetic rats,and improve the pancreas injury in a dose-dependent pattern.
