摘要
目的基于气相色谱-质谱联用技术(gas chromatography-mass spectrometry,GC-MS)对来源于黑龙江省两个稻米产地的30份稻米进行代谢组学研究。方法在R软件平台下,采用XCMS软件包对GC-MS数据进行处理,结合SIMCA-P 13.0软件,以主成分分析(Principal Component Analysis,PCA)和偏最小二乘法-判别分析(Partial Least Squares-Discriminate Analysis,PLS-DA)进行多元统计分析。结果共检测出64种包括糖、氨基酸、脂肪酸及有机酸等代谢产物,对显著变化(P<0.05,VIP≥1)的代谢物进行鉴定,筛选出11种差异代谢物,可作为稻米产地区分的关键指标,表明稻米的代谢产物携带其产地信息,进一步为靶向代谢组学提供准确研究指标。结论利用此方法分析环境对水稻代谢产物的影响具有一定的可行性,为因环境不同而导致的稻米品质差别提供了可视化的理论基础,也间接为稻米产地鉴别提供了技术支持。
Objective To investigate the metabonomics of 30 rice samples from two rice-producing areas in Heilongjiang Province,China based on the technique of gas chromatography-mass spectrometry(GC-MS). Methods The GC-MS data were analyzed by XCMS software package under the R software platform. Combined with SIMCA-P 13. 0 software,multivariate statistical analysis was carried out by using the Principal Component Analysis(PCA) and the Partial Least Squares-Discriminate Analysis(PLS-DA). Results A total of 64 kinds of metabolites including sugar,amino acids,fatty acids and organic acids were detected. Eleven kinds of differential metabolites were screened from the metabolites that have been identified with significant changes(P〈 0. 05,VIP ≥ 1),which might be used as a key index to distinguish the rice producing areas,which indicated that the metabolites of rice carried the information of its origin and further provided accurate indicators for study on targeted metabolomics. Conclusion It is feasible to analyze the effect of environment on rice metabolites,which provides a theoretical basis for visualization of difference in rice quality due to different environments as well as a technical support for rice origin identification.
