肺结核患者医院感染后外周血miR-29a-3p水平与巨噬细胞因子的相关性研究 被引量:5

Correlation between the level of miR-29a-3p and the expression of macrophages released cytokines in the peripheral blood of tuberculosis patients with nosocomial infections

摘要 目的研究肺结核合并医院感染患者外周血miR-29a-3p水平与巨噬细胞相关因子的相关性。方法将2016年1月-2017年1月于医院结核内科住院的发生医院感染的32例LTBI患者和29例TBI患者纳入研究,将患者分为LTBI组和TBI组。抽取患者外周血提取总RNA反转录后进行qPCR反应检测miR-29a-3p表达水平。采用酶联免疫吸附实验(ELISA)法对两组患者血清中白细胞介素-6(Interleukin-6,IL-6)、白细胞介素-10(Interleukin-10,IL-10)及肿瘤坏死因子-α(Tumor necrosis factor,TNF-α)水平进行检测,Pearson法研究相关性。结果 LITB组患者外周血miR-29a-3p表达量(1.56±0.48)高于TBI患者表达水平(0.77±0.45)(t=5.599,P<0.001);LITB组患者IL-6及TNF-α水平低于TBI组患者,IL-10水平高于TBI组患者(P<0.05);Pearson相关性分析表明,LITB组与TBI组患者外周血中miR-29a-3p与IL-6及TNF-α均呈负相关关系,与IL-10呈正相关关系(P<0.05)。结论 miR-29a-3p在结核病发病具有重要作用,可能参与巨噬细胞抗结核免疫反应。 OBJECTIVE To investigate the correlation between the level of miR-29 a-3 p and the expression of macrophage-released cytokines in the peripheral blood of patients with tuberculosis infection(TBI)combined with nosocomial infection.METHODS A total of 32 patients with latent TB infection(LTBI)and 29 patients with TBI who were hospitalized in the department of tuberculosis medicine from Jan.2016 to Jan.2017 were selected,and were divided into LTBI group and TBI group.The total RNA was extracted from peripheral blood of patients and the expression level of miR-29 a-3 p was detected by qPCR reaction after reverse transcription.The levels of interleukin-6(IL-6),interleukin^-10(IL^-10)and tumor necrosis factor-α(TNF-α)in the serum of the two groups were detected by enzyme linked immunosorbent assay(ELISA).The Pearson method was related to the study.RESULTS The expression level of miR-29 a-3 p of patients of LITB group(1.56±0.48)was significantly higher than that of the patients of TBI group(0.77±0.45)(t=5.599,P〈0.001).The peripheral levels of IL-6 and TNF-αof patients of LITB group were significantly lower than those of patients of TBI group(P〈0.05).Pearson assay revealed that the correlations between the level of miR-29 a-3 p and the expressions of IL-6 and TNF-αin patients of LITB group and those of TBI group were negative,and which was positive between the level of miR-29 a-3 p and the expressions of IL^-10(P〈0.05).CONCLUSION miR-29 a-3 p might play an important role in the infection and pathogenesis of pulmonary tuberculosis,and might involved in the anti-TB immunoreaction of macrophages.
