羊膜泪道支架预防围绝经期雌兔干眼症

Experimental study on prevention of dry eye in peri-menopausal female rabbits with amniotic lacrimal duct stent

目的探究羊膜泪道支架对围绝经期雌兔干眼症的预防效果。方法选取36只健康雌兔,切除双侧卵巢,随机分为3组(每组12只),其中:A组为阴性对照组,不进行任何处理,B组为假植入组,C组为羊膜泪道支架预防组,并在植入羊膜泪道支架前、植入后2、4、6 w分别对A、B、C 3组进行泪液分泌实验(SIT)、角膜荧光素染色检查(FL)、光学相干断层扫描血管造影(OCTA)和角膜共聚焦显微镜检查。结果羊膜抗拉强度为(0.691±0.221)MPa,断裂伸长率为(19.021±4.321)%。干预后2、4、6 w 3组泪液分泌量均显著少于干预前(均P<0.05),FL评分均显著高于干预前(均P<0.05);C组干预后泪液分泌量显著多于同时段A组及B组(均P<0.05),FL评分均显著低于同时段A组及B组(均P<0.05);6 w后,C组上皮基底细胞密度及炎症细胞密度均显著少于A组及B组(均P<0.05);C组各区域角膜上皮厚度及全层厚度均明显比A、B组薄(均P<0.05),角膜上皮的ST5、S5、SN5、ST6、S6、SN6区域显著比M区薄(均P<0.05),角膜全层的T5、IT5、IN5、N5、T6、N6区显著比M区角膜厚(均P<0.05)。结论植入羊膜泪道支架对围绝经期雌兔干眼症的预防有一定效果。

Objective To investigate the preventive effect of amniotic lacrimal duct stent on dry eye syndrome in peri-menopausal female rabbits. Methods 36 healthy female rabbits were removed bilateral ovaries and randomly divided into 3 groups（ 12 in each group）.Among them,group A was negative control group and no treatment was performed. Group B was pseudo-implantation group,group C was the amniotic lacrimal duct stent prevention group. The tear secretion test（ SIT）,corneal fluorescein staining（ FL）,optical coherence tomography angiography（ OCTA） and corneal confocal microscopy were performed in groups A,B,and C before and after implantation of amniotic lacrimal duct stents. Results The tensile strength of amniotic membrane was（ 0. 691 ± 0. 221） MPa and the elongation at break was（ 19. 021±4. 321） %. The amount of tear secretion of 3 groups in the 2,4,and 6 w was significantly lower than that of before the intervention（ all P〈0. 05）,and the FL scores were significantly higher than those of before the intervention（ all P〈0. 05）. The tear secretion was significantly higher in the C group than that in group A and group B during the same period（ both P〈0. 05）,the FL scores were significantly lower than those in group A and group B during the same period（ both P〈0. 05）. After 6 weeks,the epithelial basal cell density and inflammatory cell density in group C were significantly less than those in group A and B（ both P 0. 05）; corneal epithelial thickness and full thickness were significantly thinner in group C than in group A and B（ both P〈0. 05）. The SN6 region of corneal epithelial was significantly thinner than the M region（ both P〈0. 05）. The T5,IT5,IN5,N5,T6,and N6 regions of the full-thickness cornea were significantly thicker than those in the M region（ P〈0. 05）. Conclusions Implantation of amniotic lacrimal duct stent has a certain effect on the prevention of dry eye syndrome in peri-menopausal female rabbits.
