巴西橡胶树逆境相关HbCPK1结构与功能分析

Structure and Function Analysis of HbCPK1 Relevant to Stress in Rubber Tree(Hevea brasiliensis Muell. Arg.)

摘要 钙依赖型蛋白激酶(calcium-dependent protein kinases,CDPK,CPK)在植物对生物和非生物胁迫抗性中具有重要作用。为揭示橡胶树CPK家族成员在橡胶树白粉病抗性中的作用,从巴西橡胶树‘热研7-33-97’叶片中克隆了一个CPK基因,其推导氨基酸含有特征性蛋白激酶家族结构域和4个EF手型蛋白结构域,命名为HbCPK1。该基因在花、叶片、胶乳和树皮中均有表达,在花中表达量最高。干旱、机械伤害和白粉病浸染均显著上调该基因在橡胶树叶片中表达。表明HbCPK1主要受非生物胁迫反应诱导,参与橡胶树对逆境响应,本研究为阐明HbCPK1基因在橡胶逆境胁迫响应下的作用提供理论参考。 The calcium-dependent protein kinases(CDPK,CPK) play an important role in biotic and abiotic stress resistance in plant.In this study,a CPK gene named HbCPK1 was cloned from leaves of rubber tree 'CATAS7-33-97' to reveal the functions of CPK gene family in rubber tree powdery mildew resistance.HbCPK1,containing specific protein kinase family domain and 4 EF-hand protein domains,was expressed in bark,flower,leaf and latex,which had the highest expression level in flower.Drought,mechanical wounding and powdery mildew infection up-regulated its expression significantly in rubber leaves.These suggested that HbCPK1,inducted by abiotic stresses,played roles in stress responses of rubber tree.This study might provide a theoretical reference for clarifying the role of HbCPK1 gene in response to stress of rubber.
