摘要
玉米的正反交杂交种在有些表型性状上存在差异,在有些品种中甚至直接影响总体产量。为了避免之前的鉴定正反交方法在取样类型和时间上存在的一定的局限性,本研究以玉米叶绿体基因组Insertion/Deletion(In Del)标记为基础开发引物,使用玉米叶片材料进行检测。结果显示CPMIDP01和CPMIDP03两个位点可分别鉴定玉米杂交种郑单958和京科968的正反交组合,并可组合成效果稳定的二重PCR,从而得到一种对玉米正反交种的快速鉴定方法。此方法可应用于中国主要玉米品种正反交鉴定中,作为核基因组检测的补充,进一步完善品种真实性及品系溯源信息。
Phenotype differences were observed between some maize hybrid and reciprocal cross hybrid in some traits,especially in grain yield.The identification method of reciprocal cross reported previously had limitation especially on sample type and time.Primers were developed with leaf materials based on Insertion/Deletion(In Del) markers in the maize chloroplast genome.The results showed that CPMIDP〈01 and CPMIDP〈03 were identified maize hybrid and reciprocal cross Zhengdan958 and Jingke968 separately and could be combined into2-plex PCR stably.As a result of establish a rapid identification method for maize normal and reciprocal cross hybrids.This method can be applied to identify of normal and reciprocal cross of main maize varieties in China,as a supplement for identification nuclear genome,further improves variety authenticity and traceability information.
