四种分子标记技术在甘薯及I.trifida遗传多样性研究中的应用 被引量:4

摘要 本研究利用ISSR以及在甘薯中报道较少的3种分子标记技术SCoT、CDDP、CBDP,分别对46份甘薯及其近缘属I.trifida进行基因组扫描。结果表明,SCoT、CDDP、CBDP 3种分子标记的单引物平均扩增片段数和扩增多态性均优于目前甘薯研究中常用的ISSR分子标记。4种标记分析结果遗传相似性系数变异范围0.36~0.92,各分子标记获得的遗传相似性系数差异不大。利用全部分子标记结果进行聚类分析和主坐标分析表明,参试材料之间差异较明显。甘薯与I.trifida间遗传多样性丰富,遗传差异较大。在不同甘薯品种之间,除少部分材料间遗传差异较大外,大部分甘薯品种间的差异较小。本研究结果初步证明SCoT、CDDP、CBDP 3种分子标记可应用于今后甘薯及其近缘属分子标记研究,更好的发掘甘薯及其近缘属的遗传多样性。此外,利用I.trifida与栽培甘薯较大的遗传差异,丰富栽培甘薯遗传多样性,将对甘薯育种发展具有重要作用。 In the present study,four molecular marking techniques,containing ISSR,SCoT,CDDP,CBDP,were used to analysis genetic diversity of 46 genotypes of sweetpotato and its related species I.trifida.The results indicated that the average amplified fragment number and amplification polymorphism of single primer of SCoT,CDDP and CBDP were better than ISSR which was commonly used in the study of sweet potato at present.The genetic similarity coefficient of four marker analysis ranged from 0.36 to 0.92,which showed that there was no great difference among four marking techniques.Cluster analysis and element analysis with all marking results suggested that 46 genotypes had distinctive features.Furthermore,the genetic diversity between sweetpotato and I.trifida was very abundant and the genetic difference was big.Except for a few genetic differences among different sweet potato varieties,most varieties of sweet potato had little difference.These results preliminarily demonstrated that SCoT,CDDP and CBDP could be further applied in the molecular marker analysis of sweetpotato and I.trifida,so as to explore the genetic diversity of sweetpotato and I.trifida.Additionally,the genetic difference between sweetpotato and I.trifida could enrich the genetic diversity of cultivated sweetpotato,which would play an important role in the development of sweetpotato breeding.
