食品中4种生育酚异构体的超高效液相色谱-大体积流通池荧光测定法

摘要 目的建立具有大体积流通池的荧光检测超高效液相色谱法测定食品中4种生育酚异构体的方法。方法样品中的生育酚异构体经皂化和液液萃取,Waters BEH Shield RP18色谱柱(2.1 mm×150 mm,1.7μm)分离,以乙腈加甲醇(1∶1)及纯水作为流动相,梯度洗脱,采用具有大体积流通池的荧光检测器检测。结果 4种生育酚异构体1~60μg/ml范围内呈良好的线性关系(r2≥0.999 8),平均回收率为78.8%~111%,相对标准偏差(RSD)为0.22%~10.8%。结论方法简便、快速、灵敏度高、试剂用量少,适用于食品中生育酚的常规检测。 [Objective]To establish a method for the determination of 4 isomers of tocopherol in foods by ultra-performance liquid chromatography(UPLC) with large volume flow cell. [Methods] The tocopherols in samples were conducted liquid-liquid extraction after saponification,and then separated by Waters BEH Shield RP18 column(2.1 mm×150 mm,1.7 μm). Chromatographic mobile phase was methanol,acetonitrile(1 ∶1) and water and by gradient elution. The target analyte was detected by the fluorescence detector with large volume flow cell. [Results] The linear ranges of tocopherols were from 1-60 μg/ml(r^2≥0.999 8). The average rates of recovery were from 78.8%-111% and the relative standard deviations(RSD) were from 0.22%-10.8%. [Conclusion] This method is applicable to determination of tocopherol in foods for the advantages of simple,fast,high sensitivity,low dosage of reagent.
