白芨水提物对小鼠黑色素瘤B16F10细胞增殖与迁移的影响 被引量:3

摘要 目的探讨白芨水提物对小鼠黑色素瘤B16F10细胞增殖与迁移的影响。方法使用水提醇沉法提取白芨粗多糖,同时保留有机小分子物质部分;采用CCK8试剂检测不同浓度的两部分提取物对B16F10细胞存活率及增殖的影响;细胞划痕检测提取物对B16F10细胞迁移的影响,以及ELISA法检测迁移相关蛋白的表达情况。结果与对照组相比,白芨水提物均能降低B16F10细胞的存活率和增殖能力,并与浓度呈正相关,其中多糖和有机小分子物质的半数抑制浓度(IC_(50))分别为51.14μg/mL和40.61μg/mL;此外,白芨水提物对细胞迁移也具有抑制作用,并能下调MMP-2和MMP-9的表达,与对照组相比,中、高浓度的下调作用具有显著性差异。结论白芨水提物能够抑制B16F10细胞的增殖与迁移能力,其作用可能通过下调迁移相关蛋白MMP-2和MMP-9的表达有关。 Objective To explore the effect of aqueous extract of Bletilla striata on proliferation and migration of mouse melanoma B16F10 cells. Methods Crude polysaccharides of Bletilla striata were extracted by aqueous extraction and alcohol precipitation,while the part of organic small molecule was preserved. After treatment with different concentrations of extracts,the cell viability and proliferation were detected by CCK8 assay,the cell migration was determined by cell scratch,as well as the expressions of migration-related proteins were detected by ELISA assay. Results Compared with control group,B16F10 cell viability and proliferation were reduced in a dose-dependent manner,and the IC50 values of polysaccharide and organic small molecule were 51. 14 μg/mL and 40. 61 μg/mL,respectively. In addition,the cell migration activity was inhibited and the expressions of MMP-2 and MMP-9 proteins were down-regulated after cultured with aqueous extract of Bletilla striata. Compared with control group,the down-regulation effects of medium and high concentration groups were significantly different. Conclusion The aqueous extract of Bletilla striata can inhibit the proliferation and migration of B16F10 cells,which may be related to the down-regulation of the protein levels of MMP-2 and MMP-9.
