摘要
目的:探讨丹参酮Ⅱ_A促食管癌细胞系Eca109细胞凋亡的可能机制。方法:Eca109细胞应用不同浓度(2、4、8μg/mL)丹参酮Ⅱ_A处理48 h后,应用荧光定量实时PCR及Western blotting分析凋亡相关蛋白BAX、Bcl-2、Caspase-3、Caspase-9、GRP78、XIAP mRNA及蛋白的表达。结果:丹参酮Ⅱ_A可促进Eca109细胞凋亡,抑制Bcl-2、XIAP、GRP78 mRNA及蛋白表达,促进BAX、Caspase-9、Caspase-3 mRNA及蛋白表达,其作用呈浓度依赖性。结论:丹参酮Ⅱ_A可能通过调节凋亡相关蛋白表达促进Eca109细胞凋亡。
Objective: To investage the possible mechanisms of Tanshinone ⅡA promoting esophageal squamous cells( Eca109 cells) apoptosis.Methods: The human esophageal squamous cells( Eca109 cells) were treated with Tanshinone ⅡA for 48 h at different concentrations( 2,4,8 g/mL). The expression of apoptosis related proteins BAX,Bcl-2,Caspase-3,Caspase-9,GRP78,XIAP mRNA and protein expression were analyzed by real-time quantitative PCR( q PCR) and Western blotting. Results: Tanshinone ⅡA inhibited the cell proliferation and induced apoptosis in Eca109 cells. The mRNA and protein expression of Bcl-2,XIAP,GRP78 were inhibited,the mRNA and protein expression of BAX,Caspase-9 and Caspase-3 were increased,which in a concentration dependent manner. Conclusion: Tanshinone ⅡA inhibits the growth of esophageal squanous cells Eca109 and induces apoptosis possibly by suppressing cell apoptosis-related XIAP and GRP78 protein expression.
