摘要
目的探讨百里醌对人乳腺癌细胞株MCF-7的影响。方法应用CCK-8法检测百里醌对MCF-7人乳腺癌细胞株MCF-7的增殖抑制作用;流式细胞术检测MCF-7细胞凋亡情况。建立裸鼠乳腺癌皮下移植瘤模型,原位末端标记(TUNEL)法检测肿瘤组织细胞的凋亡情况;采用免疫组化方法检测肿瘤组织中ki-67、survivin的表达。结果百里醌可显著抑制体外人乳腺癌细胞株MCF-7生长,并明显诱导细胞凋亡;百里醌可显著抑制裸鼠乳腺癌皮下移植瘤生长,且明显降低乳腺癌肿瘤组织中Ki-67和survivin的阳性表达。结论百里醌可显著抑制人乳腺癌生长,该作用可能通过抑制Ki-67和survivin表达而实现。
Objective To explore the effect of thymoquinone on human breast cancer cell line MCF-7 in vitro and vivo. Methods The proliferation of MCF-7 human breast cancer cell line MCF-7 was detected by CCK-8 method.The apoptosis of MCF-7 cells was detected by flow cytometry. A nude mouse model of breast cancer was established in nude mice. The apoptosis of tumor cells was detected by TUNEL. The expression of ki-67 and survivin in tumor tissues was detected by immunohistochemistry. Results Thymoquinone could significantly inhibit the growth of human breast cancer cell line MCF-7 and induce apoptosis in vitro. Thymoquinone could significantly inhibit the growth of subcutaneous xenografts in nude mice, and significantly reduce Ki-67 in breast cancer tissues and positive expression of survivin. Conclusions Thymoquinone can significantly inhibit the growth of human breast cancer, which may be achieved by inhibiting the expression of Ki-67 and survivin.
