miR-370靶向作用TRAF-4对肺癌细胞增殖和凋亡的影响 被引量:3

摘要 目的探讨mi R-370在非小细胞肺癌中的表达及其对肺癌细胞增殖和凋亡的影响。方法采用RT-PCR法检测肺癌组织和细胞中miR-370的表达水平,CCK-8法检测不同转染组肺癌细胞增殖能力,流式细胞术检测肺癌细胞凋亡;双荧光素酶法及Western blot验证miR-370与肿瘤坏死因子受体相关基因4(TRAF-4)的作用关系。结果 miR-370在非小细胞肺癌组织和细胞中表达量均显著下调(P<0.01),转染miR-370 mimics后肺癌细胞A549的生长活性受到显著抑制(P<0.01),且其凋亡水平显著升高(P<0.01);生物信息学预测miR-370的靶基因为TRAF-4,双荧光素酶及Western blotting证实miR-370可靶向抑制TRAF-4蛋白的表达(PP<0.05)。结论miR-370可抑制肺癌细胞的增殖能力,并促进其凋亡,其可能的机制是靶向作用于TRAF-4蛋白。
