摘要
目的观察白术、防风、白术/防风药对对感染后肠易激综合征(PI-IBS)大鼠的作用,获得白术/防风药对协同保护肠机械屏障的证据并研究其机制。方法采用福氏痢疾杆菌灌胃法建立PI-IBS大鼠模型,评价白术、防风以及白术/防风药对的止痛、止泻效应;采用酶联免疫吸附法(ELISA)检测大鼠结肠炎症因子IL-6、IL-8的水平;采用Western Blot检测大鼠结肠黏液蛋白Mucin-1的表达水平;采用免疫荧光法检测大鼠结肠Occludin、Claudin-1紧密连接蛋白的表达变化。结果白术、防风以及白术/防风药对均可不同程度地升高PI-IBS大鼠的内脏疼痛(抬腹和拱腹)压力阈值,减少稀便次数(P <0. 05);白术/防风药对可以协同降低PI-IBS大鼠肠道IL-6、IL-8炎症因子水平(P <0. 05),增加结肠黏液蛋白Mucin-1的表达(P <0. 05),上调Occludin、Claudin-1紧密连接蛋白的表达。结论白术、防风、白术/防风药对对PI-IBS大鼠肠机械屏障具有不同程度的保护作用,且白术防风合用起协同增效作用,其对肠机械屏障的保护机制可能是通过降低相关炎症因子IL-6、IL-8的水平,上调黏液蛋白Mucin-1、Occludin、Claudin-1紧密连接蛋白的表达实现。
Objective To investigate the effect of Atractylodes macrocephala( AM), Saposhnikovia divaricate(SD) and com patibility of AM with SD on post - infectious irritable bowel syndrome ( PI - IBS ) rats, and reveal the underlying mechanism Methods Rat models of PI - IBS were established. The therapeutic effects of AM and SD on PI - IBS rat were evaluated by measuring the rat visceral pain pressure threshold and recording the number of tools. The levels of IL - 6 and IL - 8 were meas- ured by ELISA. The expression of Mucin - 1 protein was analyzed by western blot. The levels of the tight juction Oecludin and Claudin - 1 proteins were examined by immunofluorescence. Results AM, SD and compatibility of AM with SD could enhanced the rat visceral pain pressure threshold and decrease the number of stools (P 〈 0. 05 ). AM, SD and compatibility of AM with SD could decrease the level of IL- 6 and IL- 8 (P 〈 0. 05), increase the expression of Mucin- 1 protein( P 〈 0. 05 ), improve theexpression of Oeeludin and Claudin - 1 protein in different degrees. Conclusion AM, SD and the eompatibility of AM with SD has a the rapeutic effect on intestinal barrier in different degree, and the compatibility of AM with SD has a synergistic effect. The mecha- nism may be related to the reduce of IL - 6, IL - 8 level, theup - regulation of the expression of Muein - 1 protein, Oeeludin and Claudin - 1 protein.
