骨髓增生异常综合征患者表观遗传学改变的研究进展

Advances of epigenetic changes in patients with myelodysplastic syndrome

摘要 骨髓增生异常综合征(MDS)是一类具有临床和遗传学异质性的骨髓衰竭性疾病.表观遗传学改变是指基因表达发生克隆性的可遗传改变,但是不伴有核苷酸序列的改变,主要包括DNA甲基化、组蛋白修饰及非编码RNA调节.近年,实验室及临床研究结果均表明,表观遗传学改变在MDS的发生、发展中发挥至关重要的作用,并且对其治疗创新及进展发挥关键作用.笔者拟就DNA甲基化、组蛋白修饰及非编码RNA调节等表观遗传学改变在MDS发病及治疗中的研究进展进行综述. Myelodysplastic syndrome (MDS) is a group of bone marrow failure diseases with clinically and genetically heterogeneous.Epigenetic changes refer to the cloned and heritable changes in gene expression,but not with the changes of nucleotide sequence,which mainly include aberrant methylation of DNA,histone modification and non-coding RNA regulation,etc..In recent years,laboratory and clinical researches showed that epigenetics played an important role in the pathogenesis of MDS,and brought the innovation of MDS treatment.This article mainly reviews the effects of epigenetic changes such as DNA methylation,histone modification and non-coding RNA regulation on the pathogenesis and treatment of MDS.
