利用响应面分析法优化聚乙烯醇壳聚糖水凝胶配方 被引量:2

摘要 制备聚乙烯醇壳聚糖水凝胶,运用单因素实验考察聚乙烯醇与壳聚糖的质量比(g/g)、交联剂戊二醛终浓度、冰醋酸浓度以及反应温度对水凝胶溶胀性能的影响;应用响应面分析法对聚乙烯醇壳聚糖水凝胶的制备方法进行优化;通过糖尿病大鼠足溃疡模型验证水凝胶对糖尿病溃疡创面愈合的影响。结果单因素实验和响应面法证实在PVA︰CS(g/g)=3︰1、戊二醛终浓度为0.30 mol/L、反应温度在80℃的制备条件下水凝胶的溶胀性能达到最佳;通过糖尿病大鼠足溃疡面愈合的验证实验表明按最佳制备方法制得的聚乙烯醇壳聚糖水凝胶可用于糖尿病大鼠足溃疡创面的治疗。 Polyvinyl alcohol chitosan hydrogel was prepared. The effects of the mass ratio of polyvinyl alcohol to chitosan(g/g), crosslinking agent glutaraldehyde final concentration, acetic acid concentration and reaction temperature on the swelling properties of hydrogels were investigated by single factor experiment. The method of preparing polyvinyl alcohol chitosan hydrogel was optimized by response surface analysis, the effect of hydrogel on wound healing of diabetic ulcer was verified by diabetic rat foot ulcer model. The results of single factor experiment and response surface method to confirm the PVA︰CS(g/g)=3︰1, glutaraldehyde concentration is 0.30 mol/L, the preparation conditions of the reaction temperature at 80 ℃ water gel swelling performance to achieve the best; The experimental results show that the polyvinyl alcohol chitosan hydrogel prepared by the best preparation method can be used in the treatment of diabetic rat foot ulcer wound.
