摘要
目的探讨血清miRNA-6767-5p在多囊卵巢综合征患者中的表达及临床意义。方法选取120例多囊卵巢综合症(PCOS)患者及40例正常体检妇女(对照组)为研究对象,用qRT-PCR检测受试者血清中miRNA-6767-5p水平,并测量身高、体重并计算体重指数。测定血清卵泡刺激素(FSH)、黄体生成激素(LH)、睾酮(T)、空腹血糖(FBG)、空腹胰岛素、血清性激素结合球蛋白(sex hormone-binding globuli,SHBG)等。采用HOMA-IR公式计算胰岛素抵抗指数。结果 PCOS组血糖、胰岛素,HOMA-IR,LH和雌二醇水平高于对照组,差异具有统计学意义(P均<0.05);PCOS组SHBG水平低于对照组,差异有统计学意义(P<0.001)。PCOS组miRNA-6767-5p水平[(6.28±0.98)×10~5]低于对照组[(8.05±1.02)×10~5],差异有统计学意义(t=11.47,P<0.001)。miRNA-6767-5p与SHBG呈正相关,差异具有统计学意义(t=10.03,r=0.602,P=0.042);miRNA-6767-5p与空腹血糖呈负相关,差异具有统计学意义(t=15.84,r=-0.783,P=0.021)。miRNA-6767-5p预测PCOS的AUC、灵敏度、特异性、阳性预测值、阴性预测值分别为0.714(0.601~0.827)、63.64%、40.73%、70.00%、64.25%。结论 miRNA-6767-5p可能是诊断多囊卵巢综合征的一种新的分子标志物,在PCOS代谢中可能起到一定的作用。
Objective To examine the role of the serum miRNA-6767-5p as a biomarker for the diagnosis of PCOS and its relationship with metabolic and reproductive processes. Methods 120 patients with PCOS and 40 healthy controls were included in the study. Serum miRNA-6767-5p levels were measured by enzyme-linked immunosorbent assay. The height, weight of patients were measured and then used to calculate the body mass index. The serum levels of follicle stimulating hormone ( FSH), luteinizing hormone ( LH), testosterone ( T), fasting blood glucose ( FBG ), fasting insulin ( INS ) , and Sex hormone- binding globuli ( SHBG ) were detected, and insulin resistance index was also calculated according to HOMA-IR model formula. TaqMan quantitative real- time polymerase chain reaction ( RT- qPCR) assays was used to exam the expression of the miRNA-6767-5p. Results The levels of blood glucose, insulin,HOMA-IR, LH and estradiol in PCOS group were higher than those in the control group ( all P 〈0.05 ) ; The SHBG level in PCOS group was lower than that in the control group( P 〈0. 001 ). The level of miRNA-6767-5p in group PCOS [ ( 6.28 ± 0.98 ) × 10^5] was lower than that of the control group E (8.05 ± 1.02) × 10^5] (t = 11.47,P 〈0. 001 ). miRNA-6767-5p was positively correlated with SHBG( t = 10.03, r = 0. 602, P = 0. 042 ) ; miRNA-6767-5p was negatively correlated with fasting blood glucose( t = 15.84, r = - 0. 783, P = 0. 021 ). The AUC, sensitivity, specificity,positive predictive value and negative predictive value of miRNA-6767-5p predicting PCOS were 0. 714 (0. 601 - 0. 827 ), 63.64%, 40.73 %, 70%, and 64.25 %, respectively. Conclusion Serum miRNA-6767-Sp could be a novel candidate as a molecular biomarker in the diagnosis of PCOS and may participate in the development of the metabolic and reproductive processes of PCOS.
