摘要
目的探讨乳腺癌组织中纺锤体和动粒相关蛋白2(SKA2)mRNA和蛋白的表达水平,并分析其与临床病理特征的相关性。方法收集新鲜乳腺癌及癌旁冷冻组织标本40对,采用实时荧光定量PCR法检测SKA2 mRNA表达水平;选取乳腺癌石蜡包埋标本160例,采用免疫组化染色法检测SKA2蛋白表达水平,并分析SKA2蛋白表达水平与患者年龄、肿瘤大小、TNM分期、组织学分级、病理分型、分子分型及淋巴结转移状况等临床病理特征的相关性。结果乳腺癌组织中SK2 mRNA表达水平明显高于癌旁组织,差异有统计学意义(P<0.05)。SKA2 mRNA在乳腺癌中的高表达率为70.0%。SKA2蛋白表达与患者TNM分期和淋巴结是否转移均有关(均P<0.01),而与患者年龄、肿瘤大小、组织学分级、病理分型、分子分型均无关(均P>0.05)。结论SKA2蛋白在乳腺癌中的高表达与TNM分期和淋巴结转移密切相关;SKA2可作为反映乳腺癌转移的一个重要指标。
Objective To investigate the expression of spindle and kinetochore associated protein 2 (SKA2) in breastcancer tissues and analyze the relationships between SKA2 expression and clinicopathological factors. Methods The relativeSKA2 mRNA expression was detected by real time quantitative polymerase chain reaction (Real-time qPCR) in 40 fresh or frozensamples of breast cancer and corresponding paracancerous tissue. The SKA2 protein expression was also examined byimmunohistochemistry in 160 specimens of paraffin-embeded breast cancer tissues. The correlation of SKA2 with theclinicopathological characteristics including age, tumor size, clinical stage, histological grade, pathological type, molecular typeand lymph node metastasis was analyzed by SPSS 18.0. Results The expression levels of SKA2 were significantly increased inbreast cancer compared to the normal control(P〈0.05). The expression of SKA2 was correlated with TNM staging and lymphnode metastasis (both P〈0.01); however, no correlated with age, tumor size, clinical stage, histological grade, pathological typeand molecular type(all P〉0.05). Conclusion High expression of SKA2 in breast cancer is closely related with the stage of tumorand lymph node metastasis; the expression of SKA2 may be used as the potential pre-metastatic marker of breast cancer in thefuture.
