摘要
目的:通过检测奶牛CD4基因选择性剪接,为研究CD4基因的结构和功能打下基础。方法:采集奶牛尾静脉血,提取血液总RNA后反转录成cDNA。通过反转录PCR扩增和测序,以鉴定CD4基因剪接体和单核苷酸多态性。结果:通过序列比对分析,发现奶牛CD4基因外显子8处存在缺失,扩增的两种CD4mRNA序列长度相差122bp(整个外显子8缺失)。另外,在牛CD4基因外显子8处检测到1个碱基突变(C/A)。结论:奶牛CD4基因发现两种相差外显子8的剪接体,并检测到1个SNP。
Objective:The bovine CD4 gene was spliced selectively,which can lay a foundation for investigating the structure and function of CD4 gene.Methods:The tail vein blood of the cow was collected,and the total RNA of the blood was extracted and reverse transcribed into cDNA.The CD4 gene splicing isoforms and single nucleotide polymorphisms(SNPs)were identified by reverse transcription PCR amplification and sequencing.Results:The deletion of exon 8 of the bovine CD4 gene was found by sequence alignment analysis,and the length difference between the two amplified CD4 mRNA sequences was 122 bp(deletion of the whole exon 8).Besides,a base mutation(C/A)was detected at exon 8 of bovine CD4 gene.Conclusion:The difference of two splicing isoforms in exon 8 were found and one SNP was detected in the CD4 gene of dairy cattle.
