摘要
目的:研究不同浓度的栽培黑豆和野生黑豆粉末对深层发酵灵芝菌丝生长、灵芝酸产量的影响。方法:通过菌丝活化、种子液培养、液体深层发酵培养3阶段获得灵芝菌丝体,用乙醇提取灵芝酸,香草醛法测定其含量并计算产量。结果:当培养基栽培黑豆浓度为10g/L时,灵芝酸含量和产量分别比对照提高了1.59和2.66倍;当培养基野生黑豆浓度为10g/L时,灵芝菌丝体生物量和灵芝酸产量分别比对照提高2.25和3.89倍;当培养基野生黑豆浓度为4g/L时,灵芝酸含量达到最大值39.845mg/g。当培养基中野生黑豆浓度为6、8、10g/L时试验组灵芝酸产量无显著差异。结论:从经济性角度考虑,添加4g/L野生黑豆优化培养基效果最佳。
Objective:Effects of black soybean powder on the mycelial growth,the content of ganoderic acid and the production of ganoderic acid were explored in submerged fermentation broth containing different concentrations of wild black soybean and cultivated black soybean powder.Methods:The Ganoderma lucidum was obtained through three stages of the mycelia activation,seed culture and submerged fermentation in vanillic aldehyde assay,and then the obtained mycelia was puritied with ethanol.Results:It showed that the content and yield of ganoderic acid in mycelia were up-regulated by 159%and266%,respectively,compared with the control group where added 10 g/L cultivated black soybean in culture solution.The mycelia biomass of Ganoderma lucidumand ganoderic acid content treated with 10 g/L wild black soybean were enhanced by 225% and 389%,respectively.The content of ganoderic acid in mycelia reached peak,being 39.845 mg/g,when 4 g/L of wild black soybean was added to medium.However,there is no significant difference in the yield of ganoderic acid between 6,8 and 10 g/L of wild black soybean.Conclusion:From the viewpoint of economy,the highest production of ganoderic acid was 4 g/L wild black soybean in media,which was selected to apply to further application.
