“502”胶熏显指印后5种脱落细胞转移方法对STR分型影响观察 被引量:2

Observation and comparative effection on developing potential fingerprints by “502” glue among five transfer methods in cast-off cells

摘要 目的黑色塑料袋上潜在指印经"502"胶熏显后,观察比较5种脱落细胞转移方法对STR分型影响。方法在黑色塑料袋上捺印手印230枚,经"502"胶熏显后分别采用实物剪取、去离子水、无水乙醇、丙酮、解胶剂等5种方法转移脱落细胞,硅珠法抽提DNA,Amp FlSTR~ Identifiler~ Plus试剂盒进行STR复合扩增,3500XL遗传分析仪电泳检测扩增产物,等位基因分型用GeneMapper~ ID-X软件判读分析。记录每个样本检出等位基因的个数。各组间差异利用SPSS13.0软件进行统计学分析。结果 5种方法转移后的检材STR检出率差异明显。以STR检出≥13个基因座为标准,去离子水组最高(检出率82.6%),解胶剂组次之(检出率78.3%),实物剪取组最低(检出率34.8%),以每例样品RFU值≥5000为检验满意标准,解胶剂组的检验满意度最高(26.1%)。结论使用解胶剂类溶剂可以有效转移"502"胶中的脱落细胞,为"502"胶熏显客体中的脱落细胞的STR检验提供新思路。 Objective To Observe,record and compare the effection on developing potential fingerprints by "502" glue among five transfer methods in cast-off cells.Methods 230 fingerprints pressed on black plastic bags were developed by "502" glue evaporation,and cast-off cells in fingerprints were transferred by clipping,deionized water,absolute alcohol,acetone and dispergator.DNA was extrated by silicon bead method,STR multiplex amplification was amplified by Amp FSTRIdentifilerPlus Kit,fluorescent multiplex amplification products were detected with 3500 XL genetic analyzer and alleles were typed with Gene MapperID-X software.The number of alleles detected per sample was recorded and the difference between groups was analyzed by SPSS13.0 software.Results There was significant difference on ratio of successful detection of biological evidence among five transfer methods.The ratios of successful detection were 82.6% in deionized water group which was the highest,78.3% taking second place in dispergator group and the lowest only 34.8% in clipping group.The highest satisfaction rate which was 26.1% appeared in dispergator group by the standard of RFU more than 5000 value for each sample.Conclusion Before DNA typing,it is effective to use solvent such as dispergator for transferring cast-off cells in "502" glue,and it provides a new idea for STR profiling of cast-off cells in "502" glue.
