摘要
为了探讨围生期蒙古牛外周血中性粒细胞(PMN)CD11b的动态表达和血清主要类固醇激素浓度变化,试验选择经同期发情处理的经产蒙古牛10头(2胎或3胎),于分娩前22,18,14,10,8,6,4,2天,分娩当天(记为0天),分娩后2,4,6,8,10,14天进行颈静脉采血,分离PMN、血清,采用RTPCR、ELISA、放射免疫(RIA)等方法测定中性粒细胞雌激素受体、CD11b mRNA表达、血清CD11b浓度及类固醇激素浓度。结果表明:PMN数量分娩前显著增加,分娩当天数量最低;产前孕酮(P4)含量逐渐下降,而雌二醇(E2)含量产前2天明显升高;雌激素受体(ERα)变化规律与雌二醇相似,但ERβ无明显变化;CD11b mRNA及血清CD11b浓度产前均明显增加,随后逐渐降低。
The aim of the present study was to investigate the dynamic expression of CD 1 l b in peripheral neutrophils of Mongolian cattle (PMN) during pefinatal period and the variation of serum main steroid hormone. The estrus managed by synchronization ten multiparity ( 2- 3 fetus) Mongolia cattle were selected in this experiment, and then PMN and serum were isolated after collecting blood on 22, 18, 14, 10, 8, 6, 4 and 2 days before delivery (denoted as-22 d,-18 d,-14 d,-10 d,-8 d,-2 d), delivery day (0 d), 2, 4, 6, 8, 10 and 14 days after delivery through jugular vein, then the expression of ERa, ERfl, CD1 l b mRNA in PMN and the concentration of CD11 b, E2 and P4 in serum were de- tected by RT-PCR, ELISA and RIA, respectively. The results showed that the amount of PMN increased significantly before delivery, and the minimum was on the day of delivery. The content of progesterone (P4) decreased gradually, while the content of estradiol (E2) increased sig- nificantly on the prenatal 2 days. The changes of ERa were similar to that of E2 , but ERI3 did not change significantly. The concentration of CD1 lb mRNA and serum CD1 lb increased significantly before delivery, and then decreased gradually.
