中国-印尼积雪草药材化学成分对比研究

Study on the Comparison of Chemical Compositions in Centella asiatica( L. ) Urb. from China and Indonesia

目的比较研究中国和印尼积雪草药材化学成分的差异。方法以积雪草苷为对照品,采用薄层色谱法(TLC)、高效液相色谱法(HPLC)及高效液相色谱蒸发光散射(HPLC-ELSD)指纹图谱对来源于中国和印尼的积雪草药材进行比较研究。结果薄层色谱鉴别:来源于中国的积雪草药材365 nm处检视有一特有的蓝色荧光斑点,两个不同来源的积雪草药材其他斑点颜色、数目及比移值(Rf值)基本一致;含量测定:来源于中国的积雪草药材中积雪草苷含量较高,达到0.52%(n=3),本批次的印尼积雪草药材中积雪草苷含量较低,为0.35%(n=3);HPLC-ELSD指纹图谱:两种不同来源的积雪草药材化学成分存在较大差异,其中1、2、5号峰为中国积雪草特有的峰,而3号(积雪草苷)、4号峰为两者共有的峰。结论首次初步建立了积雪草药材的HPLC-ELSD指纹图谱方法,有利于中医药研究的国际交流合作。

Objective To Comparatively study on the differences of chemical constituents in Centella asiatica（ L.） Urb. from China and Indonesia. Methods Asiaticoside was used as reference material to analyze Centella asiatica（ L.） Urb. from two different sources by TLC,HPLC and HPLC-ELSD-FT. Results TLC： The Centella asiatica herbs from China had a unique blue fluorescent spots under 365 nm.The color,numbers and Rf of other spots in the two different Centella asiatica were almost consistent. Assay results： The Centella asiatica herbs from China had the high content of asiaticoside with 0. 52%（ n =3）,Indonesia in a low level with 0. 35%（ n = 3）; HPLC-ELSD-FT： The chemical constituents in Centella asiatica（ L.） Urb. from China and Indonesia were quite different. The peaks of number 1,2,5 were the characteristic peaks of Centella asiatica in China,and the common peaks of number were 3,4. Conclusion The firstly establishment of fingerprint method is favor for the research and development of Chinese medicines between international exchange and cooperation.
