摘要
目的探讨从妊娠期至发育期氯化镧染毒对子代大鼠肾脏及抗氧化能力的影响。方法将32只健康成年SPF级Wistar妊娠大鼠随机分成4组,分别为对照(蒸馏水)组及2.5、5和10 g/L氯化镧染毒组,每组8只。自妊娠第0天至仔鼠出生后第21天(哺乳期),采用自由饮水方式进行染毒。断乳后至出生后第42天,仔鼠通过自由饮水方式进行染毒。分别于仔鼠出生后第14、28和42天,测定肾脏组织的谷胱甘肽(GSH)、丙二醛(MDA)的含量及谷胱甘肽过氧化物酶(GSHPx)、过氧化氢酶(CAT)、过氧化物歧化酶(SOD)的活力。结果与对照组比较,仅出生后第42天10 g/L氯化镧染毒组雄性和雌性仔鼠的肾脏GSH含量和CAT活力较低,差异均有统计学意义(P<0.05)。与对照组比较,各浓度氯化镧染毒不同时间仔鼠肾脏的GSH-Px、Cu-Zn SOD活力均下降,而MDA含量均升高,但差异均无统计学意义(P>0.05)。结论氯化镧染毒可造成仔鼠肾脏中抗氧化损伤酶类系统失衡,自由基清除能力及抗氧化能力逐渐降低,引起肾脏毒性反应。
Objective To understand the effect of exposure to lanthanum chloride from gestation to development period on tissue structure and antioxidant capacity in kidney of offspring rats. Methods Thirty-two healthy adult pregnant Wistar rats were randomly divided into four groups, eight in each, the control group(distilled water), 2.5 g/L, 5 g/L and 10 g/L lanthanum chloride exposure groups. The pregnant rats were given lanthanum chloride through drinking water from the first day of pregnancy to the 21 th day after birth of offspring(lactation period). The offspring rats were given the same water as their maternal rats from weaning to the 42 th day after birth. At the 14 th, 28 th and 42 th day after birth, the contents of glutathione(GSH), malondialdehyde(MDA) and the activities of glutathione peroxidase(GSH-Px), catalase(CAT), superoxide dismutase(SOD) in kidney tissue were measured. Results Compared with the control group, only the GSH contents and CAT activities in kidney of male and female offspring rats were lower in the 10 g/L lanthanum chloride exposure group at the 42 nd day after birth(P〈0.05). Compared with the control group, the GSH-Px and Cu-Zn SOD activities were decreased, while the MDA content was increased in kidney of offspring rats exposed to lanthanum chloride with different exposure doses during different exposure periods, but the differences were not statistically significant(P〉0.05). Conclusion Lanthanum chloride exposure may cause the imbalance of antioxidant enzyme systems in the kidney of offspring rats and reduce the ability for scavenging free radical and antioxidation gradually, resulting in kidney damage.
