摘要
目的优选红梅消总皂苷纯化工艺,并对其进行体外抗凝血实验研究。方法以红梅消总皂苷吸附率和转移率为指标,考察大孔吸附树脂型号、上样液质量浓度与上样量等因素,优化纯化工艺,再通过研究凝血酶原时间和抗凝血酶作用进行体外抗凝血实验。结果红梅消总皂苷最佳纯化工艺为采用D101型大孔吸附树脂,用2 BV(60 ml)的0.5 g/ml上样液(以原药材计),以3 BV/h的流速进行上样,静态吸附30 min,再用4 BV的50%乙醇以3 BV/h的流速洗脱。红梅消总皂苷对凝血酶原时间和抗凝血酶作用影响较大。结论优选工艺稳定可靠,可用于红梅消总皂苷纯化及抗凝血实验研究。
Objective To optimize purification technology parameters of total saponins from Rubus parvifolius L and investigate its anti-coagulant activity in vitro. Methods With the adsorption rate and transfer rate as index, purification technology is optimized by investigating the macroporous resin type, concentration of sample liquid, loading sample, et al. Anti-coagulant activity in vitro is studied with the prothrombin time of rabbit blood and antithrombin as index. Results Optimum purification technology is 2 BV sample liquid(0.5 g/ml) passed D101 resin at the rate of 3 BV/h, static adsorption 30 min, eluted by 4 BV 50 % ethanol at the rate of 3 BV/h. Total saponins from Rubus parvifolius L have great influence on prothrombin time and antithrombin. Conclusion The process is stable and reliable, which can be used for purifying and anti-coagulant experimental research.
