超声酶法对锦灯笼中黄酮和木犀草素提取工艺的优化 被引量:6

Optimization of Extraction Technology of Flavonoids and Luteolin From Calyx Seu Fructus Physalis by Ultrasonic Enzymatic

摘要 目的优选锦灯笼中总黄酮和木犀草素的提取工艺。方法以超声波辅助纤维素酶进行提取,研究酶解时间、料液比、酶解温度、酶用量等4个因素对锦灯笼总黄酮提取率的影响。采用UV法和HPLC法,分别测定锦灯笼的总黄酮和木犀草素的含量。结果超声波辅助酶法提取锦灯笼中总黄酮的最佳工艺条件:酶解时间3 h、料液比1∶10(w/v)、酶解温度60℃、纤维素酶用量75 mg/g,在此条件下得到的总黄酮和木犀草素总评分为3.951。结论该提取工艺简便、易行、高效,是提取锦灯笼总黄酮和木犀草素的可行方法,具有一定的推广价值,为进一步开发利用锦灯笼总黄酮和木犀草素提供了理论依据。 Objective To study the extraction process of total flavonoids and luteolin in Calyx Seu Fructus Physalis. Orthogonal test by L9(34), using the total flavonoids and luteolin content in the extract as the evaluation index, to optimize the extraction process. Methods Extracted by ultrasonic assisted cellulase, studied the effects of four factors on the extraction rate of total flavonoids, which included enzymatic hydrolysis time, ratio of solid to liquid, enzymolysis temperature and enzyme dosage. Adopted UV and HPLC to measure the contents of total flavonoids and luteolin in the Calyx Seu Fructus Physalis. Results The optimum conditions are as follows: ultrasonic time 3 h, ratio of solid to liquid 1∶10(w/v), ultrasonic temperature 60 ℃, 75 mg/g cellulase dosage of Calyx Seu Fructus Physalis, the total flavonoid and luteolin obtained under this condition is about 3.951. Conclusion The extraction process is simple, easy and efficient. It is a feasible method to extract the total flavonoids and luteolin from the Calyx Seu Fructus Physalis. It has a certain promotional value and provides a theoretical basis for the further development and utilization of the total flavonoids and luteolin.
