TβR1及部分SMAD家族成员在肺大疱组织中的表达及意义 被引量:1

The expression of the cytokines TβR1 and part of SMAD family members in bullae of lung and its significance

摘要 目的通过检测转化生长因子β受体1(TGF-β receptorl,TβR1)和SMAD家族中smad2、smad3及smad4在原发性自发性气胸(primaryspontaneouspneumothorax,PSP)患者肺大疱组织中的表达情况,探讨其在PSP形成过程中的作用及意义。方法收集34例PSP患者(研究组)的肺大疱组织及疱旁组织和10例对照研究对象(对照组)的正常肺组织,采用免疫组织化学染色技术检测肺大疱组织与正常肺组织中TβR1、smad2、smad3和smad4的表达情况。根据免疫组化IRS半定量评分比较研究组和对照组之间各因子的表达差异。结果(1)分别对比各细胞因子在同一PSP患者肺大疱组织和疱旁正常组织中的表达情况,TβR1(P〈0.01)、smad2(P=0.023)和smad4(P=0.015)在肺大疱组织内表达明显高于疱旁正常肺组织,差异有统计学意义。(2)研究组和对照组免疫组化结果显示,TβR1(P=0.012)、smad2(P=0.031)、smad4(P〈0.01)在PSP肺大疱组织中的表达明显高于对照组肺组织,差异有统计学意义。smad3在研究组和对照组中的表达虽然差异无统计学意义(P〉0.05),但smad3在PSP肺大疱组织中的表达量绝对值高于对照组肺组织(IRS4.253±1.719对3.260±2.213)。结论TβR1、smad2和smad4蛋白在PSP肺大疱组织中存在高表达,提示TβR1、smad2和smad4与PSP肺大疱发生、发展存在一定的相关性。 Objective This study intended to investigate the expression of TGF-β receptorl ( TβR1 ), smad2, smad3 and smad4 in bullae of lungs of PSP patients and to examine the role of these cytokines in the pathogenesis of PSP. Methods Bullae of thirty-four PSP patients who received standard video-assisted thoracic surgery were included as study group. Normal lung tissues around the bullae of lung of part of the 34 PSP specimens were used as a self-control. Normal lung tissues from ten patients without pneumothorax associated disease were used as a control. Immunohistochemical ( IHC ) staining of TβR1, smad2, smad3 and smad4 were performed on the normal lung tissue of control specimens and the bullae of lung of PSP speci- Immunoreactivity was evaluated based on immunoreactive score(IRS). Statistical analyses were performed using t-test or Fisher exact test. Results (1)Overexpression ofTβRI(P〈0.01) , smad2(P=0.023) and smad4(P =0.015) was detec- ted in the bullae of lung of PSP patients compared with normal lung tissue around the bullae of lung. ( 2 ) TβR1 ( P = 0. 012 ), smad2 (P = 0. 031 ) and smad4 (P 〈 0.01 ) was significantly high-expressed in bullae of lung of PSP patients compared with normal lung tissue of control group. The expression of smad3 in the study group and the control group was not statistically signif- icant( P 〉 0.05 ). However, the absolute value of expression of smad3 in PSP bullae tissue was higher than that in control group( IRS 4. 253 ± 1. 719 vs. 3. 260 ± 2. 213 ). Conclusion TβR1, smad2 and smad4 was significantly overexpressed in PSP lesions. These results suggest that an abnormal expression of TβR1, smad2 and smad4 may be involved in the pathogenesis of PSP.
