莪术醇联合5-氟尿嘧啶对大肠癌细胞增殖和凋亡的影响 被引量:8

Effect of Curcumol Combined with 5-fluorouracil on Proliferation and Apoptosis of Colorectal Carcinoma Cell Line Lo Vo

摘要 目的:探讨莪术醇(curcumol)联合5-氟尿嘧啶(5-Fu)对大肠癌Lo Vo细胞增殖和凋亡作用的影响,以及药物处理后,Lo Vo细胞中增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)和凋亡相关蛋白B淋巴细胞瘤-2(B cell lymphoma-2,Bcl-2)表达量的变化情况。方法:莪术醇(50 mg·L-1)单独或联合5-Fu(2 mg·L-1)作用大肠癌Lo Vo细胞,用噻唑蓝(MTT)比色法检测各组药物对细胞增殖的抑制作用,采用流式细胞术分析各组对细胞凋亡作用的影响,应用蛋白免疫印迹法(Western blot)检测各组细胞中,增殖和凋亡相关蛋白PCNA,Bcl-2蛋白表达的变化情况。结果:与空白组比较,莪术醇,5-Fu单独给药以及联合用药,均可以有效抑制大肠癌Lo Vo细胞的增殖,促进细胞凋亡(P〈0.05),同时可以降低PCNA,Bcl-2蛋白的表达(P〈0.05);与单独给药组比较,联合用药后,凋亡的促进作用更为明显(P〈0.05)。与空白组比较,各药物组可以更有效的抑制PCNA,Bcl-2蛋白的表达(P〈0.05)。结论:莪术醇联合5-氟尿嘧啶能有效抑制人大肠癌Lo Vo细胞的增殖和凋亡,与单独用药组比较,联合用药可以使癌细胞的敏感性增强,极大程度的促进人大肠癌Lo Vo细胞凋亡,且这些机制可能与下调PCNA,Bcl-2蛋白表达有关。 Objective: To investigate the effect of curcumol combined with 5-fluorouracil( 5-Fu) on the proliferation,apoptosis and the changes in the expressions of proliferating cell nuclear antigen( PCNA),B cell lymphoma-2( Bcl-2) in colorectal carcinoma cell line Lo Vo. Method: Lo Vo cells were treated with curcumol( 50 mg·L-1) and/or 5-Fu( 2 mg·L-1). The proliferation of Lo Vo cells were detected by methylthiazolyldiphenyltetrazolium bromide( MTT) assay. The apoptosis were analyzed by flow cytometer. The changes in PCNA and Bcl-2 expressions were detected by Western blot. Result: Curcumol,5-Fu and combination group could inhibit the proliferation and promote the apoptosis of Lo Vo cells. The rates of inhibition and apoptosis were significantly different from control group( P〈0. 05). And the expressions of PCNA and Bcl-2 were obviously decreased,with a significant difference from control group( P〈0. 05). After combined treatment,the promoting effect of apoptosis was more obvious than single treatment group( P〈0. 05),and the expressions of PCNA and Bcl-2 protein were significantly decreased,with a significant difference from control group( P〈0. 05). Conclusion: Curcumol combined with 5-Fu could effectively inhibit the proliferation and promote the apoptosis of Lo Vo cells. Compared with the single treatment group,the combination of curcumol and 5-Fu can increase the sensitivity of cancer cells and greatly promote the apoptosis of Lo Vo cells. These mechanisms may be related to the down-regulation of PCNA and Bcl-2 protein expressions.
