辅酶q10对子痫前期样模型大鼠TNF-α、IL-17A、MCP-1水平的影响

Intervention effect of coenzyme q10 on the changes of TNF-α, IL-17A and MCP-1 in preeclampsia-like rat models

摘要 【目的】探讨辅酶q10干预对子痫前期样模型大鼠的血压、循环内肿瘤坏死因子α(tumor necrosis factorα,TNF-α),白介素17A(interleukin-17A,IL-17A),单核细胞趋化蛋白1(monocyte chemotactic protein-1,MCP-1)和白介素1β(interleukin-1β,IL-1β)水平及以上因子在胎盘组织中表达的影响。【方法】9~10周龄孕鼠在妊娠第1天(gestation day 1,GD1)时被随机分配为子痫前期样模型组(L-NAME)、正常对照组(control)、子痫前期样模型动物+低剂量的辅酶q10预防性处理组(LQ)、子痫前期样模型动物+高剂量的辅酶q10预防性处理组(HQ)。各组大鼠在妊娠期分别连续给予辅酶q10及安慰剂。所有孕鼠进行妊娠期无创尾压测量;流式多因子检测法测定妊娠终点孕鼠血浆相关炎症因子水平;PCR测定孕鼠胎盘组织相关炎症因子m RNA表达水平变化。【结果】GD9后,L-NAME组大鼠的收缩压及舒张压与正常对照组比较差异有统计学意义(P<0.05),LQ组与HQ组的收缩压及舒张压分别明显低于L-NAME组(P<0.05);L-NAME组大鼠的血浆IL-17A、TNF-α和MCP-1水平与正常对照组比较差异有统计学意义(P<0.05),LQ组与HQ组的血浆相应因子水平显著低于L-NAME组(P<0.05);L-NAME组大鼠胎盘中IL-17A、TNF-α和MCP-1 m RNA表达水平与正常对照组比较差异有统计学意义(P<0.05),LQ组与HQ组大鼠胎盘组织中相应因子表达水平显著低于L-NAME组(P<0.05)。【结论】辅酶q10预防性干预可能通过改善由TNF-α,IL-17A和MCP-1此类炎症因子引起的炎症反应来降低子痫前期样模型大鼠的血压。 【Objective】To explore the intervention of coenzyme q10 on the changes of blood pressure, circulating tumor necrosis factor alpha(TNF-α), interleukin 17 A(IL-17 A), monocyte chemotactic protein 1(MCP-1) and interleukin 1(IL-1β) and the placenta m RNA expressions of these inflammatory cytokines in preeclampsia-like rat models.【Methods】The 9-10-week-old pregnant rats were randomly assigned to L-NAME group, normal control group, L-NAME + low dose coenzyme q10 group(LQ), and L-NAME + high dose coenzyme q10 group(HQ) on gestation day 1(GD1). Coenzyme q10 and placebo were given for 20 days respectively. All pregnant rats were subjected to noninvasive tail pressure measurements during pregnancy, and the plasma levels of inflammatory factors were determined by flow cytometry, and the m RNA expression levels of inflammatory factors in placenta tissues of pregnant rats were determined by PCR.【Results】After GD9, the systolic blood pressure and diastolic blood pressure in the L-NAME group were significantly higher than those in the normal control group(P〈0.05), which in LQ and HQ groups were significantly lower than those in the L-NAME group and the difference was statistically significant(P〈0.05). The plasma levels of IL-17 A, TNF-α and MCP-1 in the L-NAME group were significantly higher than those in the normal control group(P〈0.05), while the levels in the LQ and HQ groups were significantly lower than those in the L-NAME group with statistical difference(P〈0.05). The m RNA expression levels of IL-17 A, TNF-α and MCP-1 in the placenta of L-NAME group were significantly higher than those in the normal control group(P〈0.05),while the levels of IL-17 A, TNF-α and MCP-1 in the placenta tissues of LQ and HQ groups were significantly lower than those in the L-NAME group(P〈0.05). The placental IL-1β levels in each group were similar to those in plasma.【Conclusion】Preventive intervention of coenzyme q10 may reduce the blood pressure in preeclampsia-like model rats by improving inflammatory responses induced by inflammatory factors such as TNF-α,IL-17 A and MCP-1.
