摘要
目的研究养殖叶片山海绵Mycale phyllophila次生代谢产物,为药源海绵规模化养殖和开发提供依据。方法综合利用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、反相ODS柱色谱、半制备液相色谱等分离技术,对化学成分进行分离;结合NMR、MS等光谱方法和文献数据,鉴定化合物的结构。结果从叶片山海绵的低极性萃取物中,分离鉴定了11个单体化合物:3-吲哚甲醛(1)、(1 H-indol-3-yl)oxoacetamide(2)、3-hydroxyacetyl-indole(3)、3-(2-(1 H-indol-3-yl)-2-oxoethyl)-5,6-dihydropyridin-2(1 H)-one(4)、胆固醇(5)、麦角甾-5,24(28)-二烯-3β-醇(6)、胆甾-5,22-二烯-3β-醇(7)、花生酸(8)、1-棕榈酸甘油酯(9)、正十六烷基甘油醚(10)和对羟基苯甲醛(11)。结论所有化合物均为首次从叶片山海绵M.phyllophila中获得。
Objective To investigate the secondary metabolites of the cultured Mycale phyllophila.Methods Various chromatographic methods,such as Sephadex LH-20 chromatography,reverse phase chromatography and semi-HPLC chromatography were applied for separating compounds whose structures were determined by 1 H,13 C NMR spectral data and MS,and by comparing with data reported in literatures.Results From the low polar fraction of M.phyllophila,eleven compounds were isolated and identified as indole-3-carboxaldehyde(1),(1 H-indol-3-yl)oxoacetamide(2),3-hydroxyacetyl-indole(3),3-(2-(1 H-indol-3-yl)-2-oxoethyl)-5,6-dihydropyridin-2(1 H)-one(4),cholesterol(5),ergosta-5,24(28)-diene-3β-ol(6),cholest-5,22-diene-3β-ol(7),arachidic acid(8),1-monopalmitin(9),cetyl ethers of glycerol(10)and p-hydroxybenzaldehyde(11),respectively.Conclusion These eleven compounds were isolated from this sponge for the first time.
