针刺百会穴、风府穴对脑缺血损伤大鼠神经功能及NGF、BDNF表达的影响 被引量:14

摘要 目的:观察针刺百会穴、风府穴对脑缺血损伤大鼠神经功能及损伤侧皮质神经生长因子(nerve growth factor,NGF)、脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)表达的影响。方法:用随机数字表法将动物随机分为假手术组、模型组和针刺组,模型组和针刺组大鼠使用大脑中动脉线栓阻塞法,技术制作永久性脑缺血损伤模型;假手术组大鼠不做大脑中动脉线栓阻塞法仅做颈总动脉分离手术。针刺组取百会穴、风府穴,使用0.30 mm×25 mm毫针刺入百会穴、风府穴后,使用平补平泻捻转法作为行针手法,每次留针时间30 min,15 min后行针1次,术后4 h接受针刺治疗,每天治疗1次。每组各24只,再分为术后3 d、术后7 d、术后14 d 3个小组,每小组8只,术后4 h对大鼠进行神经功能缺损评分,运用神经缺损体征评分和感觉、运动能力评分,通过免疫组化技术进行NGF、BDNF显色,比较分析各组大鼠脑组织中阳性细胞的表达情况。结果:治疗前各组大鼠行为学功能评分无统计学意义(P>0.05),治疗后针刺组大鼠行为学功能评分与模型组比较有统计学意义(P<0.05);与假手术组比较,模型组大鼠皮质NGF、BDNF的表达显著升高(P<0.01);与模型组比较,针刺组大鼠皮质NGF、BDNF的表达显著升高(P<0.01)。结论:针刺百会穴、风府穴对脑缺血损伤大鼠神经功能有保护和修复作用,其机制可能与上调脑损伤皮质NGF、BDNF蛋白的表达有关。 Objective: To observe the influence of acupuncture at Baihui( GV 20) and Fengfu( GV 16) points on neurological function,expression of nerve growth factor and brain-derived neurotrophic factor in rats with cerebral ischemia injury. Methods:The animals were randomly divided into the sham operation group,the model group and the acupuncture group. The model group and the acupuncture group were used to make permanent cerebral ischemia injury model by the middle cerebral artery thread blocking technique. The sham operation group did not do the common carotid artery separation operation,but only received the middle cerebral artery thread blocking method. In the acupuncture group,the acupuncture took Baihui( GV 20) and Feng Fu( GV16) by yusing 00. 30 × 25 mm needle,the method of twirling with mild nourishment and mild purgation was used as the needle manipulation,until the rats were slightly trembling,30 minutes each time,moving the needle once after 15 minutes. The rats received acupuncture at four hours after the operation,and once a day. Then 24 rats in each group were divided into 3 groups again,with 8 cases in each group. Then 3 days,7 days and 14 days,4 hours respectively after the operation,the nerve function defect of rats was scored. The scores of nerve defects and sensory and motor ability were scored,and the nerve growth factor and brain derived neurotrophic factor were colored by immunohistochemical technique. The expression of positive cells in brain tissue of each group was compared. Results: There was no statistical significance( P〉 0. 05) for the behavioral function score of the rats in each group before the treatment,and the behavioral function score of the rats between the acupuncture group and the model group after the treatment was statistically significant( P〈 0. 05). Compared with the sham operation group,the expression of nerve growth factor and brain-derived neurotrophic factor in cortex of the model group increased significantly( P〈 0. 01). Compared with the model group,the expression of nerve growth factor and brain-derived neurotrophic factor in the cortex of the acupuncture group increased significantly( P〈 0. 01). Conclusion: Acupuncture at Baihui( GV 20) and Fengfu( GV 16) points can protect and repair the nerve function of rats with cerebral ischemia injury,and its mechanism may be related to the up-regulation of the expression of brain damage cortical nerve growth factor and brain derived neurotrophic factor protein.
