玛咖多糖对过氧化氢诱导的人神经母细胞瘤细胞损伤的保护作用 被引量:1

摘要 目的探讨玛咖多糖对过氧化氢诱导的人神经母细胞瘤细胞损伤的保护作用及其作用机制。方法采用体外细胞培养的方法,建立人神经母细胞瘤细胞的过氧化氢损伤模型,并给予玛咖多糖预保护。观察细胞形态,MTT法测定细胞存活率,测定培养液中乳酸脱氢酶活性,以及胞浆中超氧化物歧化酶、过氧化氢酶活力。流式细胞仪检测细胞活性氧酶含量以及细胞周期。结果与过氧化氢模型组相比,玛咖多糖给药组可明显提高细胞的存活率,减轻过氧化氢引起的人神经母细胞瘤细胞的损伤,且呈浓度依赖性关系,可减少乳酸脱氢酶的释放,提高超氧化物歧化酶、过氧化氢酶活力,缓解细胞周期S-期阻滞。结论玛咖多糖对过氧化氢诱导的人神经母细胞瘤细胞有明显的保护作用,其机制可能与提高抗氧化酶活力,减少细胞凋亡有关。 Objective To investigate the protective effect and possible mechanism of Maca polysaccharides(MP)on human neuroblastoma(SH-SY5 Y)cell injury induced by H_2O_2.Methods A model of H_2O_2 induced injury to SH-SY5 Ycells was established and pretreated with MP to test the protective effects.The morphological changes of cells were observed.The viability of cells was measured with the method of MTT.The content of LDH and the activities of SOD and CAT were detected with kits.The contentof ROS and cell cycle were detected via flow cytometry.Results Compared with H_2O_2 group,MP(25,50,100μg·ml^(-1))alleviated the damage to SH-SY5 Ycells induced by H_2O_2.The cell survival rate was increased significantly.The release of LDH was reduced,and the activities of SOD and CAT were increased.The apoptosis and ROS-related levels of cells were reduced,and the cell cycle retardation in S-phase was relieved.Conclusion MP has a significant protective effect on the injury to SH-SY5 Y cells induced by H_2O_2.The possible mechanism may be anti-oxidation and anti-apoptosis.
