IL-33与Treg细胞在微小病变型肾病发病机制中的作用

The role of interleukin-33 and regulatory T cells in the mechanisms for minmal change nephrotic syndrome

目的研究IL-33及调节性T(Treg)细胞在微小病变型肾病(MCNS)发病机制中的作用。方法采用ELISA法检测30例MCNS患者(试验组)和30例健康体检者(对照组)血清IL-33水平,流式细胞术测定外周血淋巴细胞亚群及Treg细胞比例,分析Treg细胞比例的变化以及与血清IL-33水平的关系。结果试验组外周血中CD4^+CD25^+T细胞及CD4^+CD25^+Foxp3^+T细胞比例低于对照组(P<0.05),而IL-33水平高于对照组(P<0.05)。试验组外周血CD4^+CD25^+Foxp3^+T细胞比例与24-h尿蛋白定量呈负相关(r=-0.66,P<0.05)。试验组血清IL-33水平与24-h尿蛋白定量呈正相关(r=0.85,P<0.05)。试验组外周血CD4^+CD25^+Foxp3^+T细胞比例与血清IL-33水平呈负相关(r=-0.70,P<0.05)。结论提升患者外周血Treg细胞水平和降低血清IL-33水平可能为治疗MCNS的新方法。

Objective To study the role of IL-33 and regulatory T（Treg）cells in the mechanisms for minmal change nephrotic syndrome（MCNS）.Methods Serum IL-33 level was detected in 30 cases with MCNS（group A）and 30 healthy controls（group C）by ELISA.Peripheral blood lymphocyte subsets and the proportions of Treg cells were detected by flow cytometry in both groups as well.The proportion of Treg cells and its relationship with serum IL-33 were analyzed.Results The proportions of CD4＋CD25＋T cells and CD4＋CD25＋Foxp3＋T cells in peripheral blood were lower,but serum IL-33 level was higher in group A than those in group C（P〈0.05）.The proportion of CD4＋CD25＋Foxp3＋T cells was negatively correlated with 24-h urinary protein（r=-0.66,P〈0.05）,but serum IL-33 level was positively correlated with 24-h urinary protein in group A（r=0.85,P〈0.05）.The proportion of CD4＋CD25＋Foxp3＋T cells in peripheral blood was negatively correlated with serum IL-33 level（r=-0.70,P〈0.05）.Conclusion Elevating the proportion of Treg cells in peripheral blood and reducing serum level of IL-33 may become a new treatment in the patients with MCNS.
