响应面法优化磷酸化改性花生分离蛋白膜制备工艺 被引量:2

摘要 目的研究磷酸化改性花生分离蛋白膜的制备工艺方法。方法以磷酸化改性花生分离蛋白为原料,以蛋白浓度、pH值、甘油百分含量(占蛋白)、黄原胶百分含量(占蛋白)、时间、温度、超声波功率、超声波频率为考察因素,以膜厚度、吸水率和透光率为考察指标,在单因素实验基础上,通过响应面实验设计进行工艺优化。结果磷酸化改性花生分离蛋白膜的最优制备工艺条件为蛋白浓度5.4%、p H值9.0、甘油百分含量(占蛋白)23.4%、黄原胶百分含量(占蛋白)4.2%、时间60 min、温度66℃、超声波功率210 W、超声波频率28 kHz;此工艺条件下的膜厚度、吸水率和透光率的响应面模型预测值分别为69μm、44.3%和50.7%,验证实验值分别为70±1μm、45.4%±1.6%和49.8%±1.4%,与模型预测值相差1.45%、2.48%和1.78%,说明模型与实际情况拟合较好,验证了预测模型的正确性。结论响应面法对磷酸化改性花生分离蛋白膜制备工艺条件参数优化是可行的,得到的工艺条件具有实际应用价值。 Objective To investigate the preparation technology of phosphorelated modified peanut protein isolate films. Methods The phosphorelated modified peanut protein isolate was used as raw material, the protein concentration, p H value, glycerol percentage content of protein, xanthan gum percentage content of protein, time, temperature, ultrasonic power, and ultrasonic frequency were used as investigation factors, and the film thickness, water absorption and light transmittance were used as the evaluation indexes. On the basis of single factor experiment, the process was optimized by response surface methodology and Box-Benhnken design. Results The optimum preparation process of phosphorelated modified peanut protein isolate films were: the protein concentration of 5.4%, p H value of 9.0, glycerol percentage content of protein of 23.4%, xanthan gum percentage content of protein of 4.2%, time of 60 min, temperature of 66 ℃, ultrasonic power of 210 W and ultrasonic frequency of 28 k Hz. The model predictive film thickness, water absorption and light transmittance under these process conditions were 69 μm, 44.3% and 50.7%, respectively. The values of verification experiment were 70±1 μm, 45.4%±1.6%% and 49.8%±1.4%, respectively. And the differences between the predicted values and the experimental values were 1.45%, 2.48% and 1.78%, respectively. The model was fitted well with the actual situation and the correctness of the proposed model was verified. Conclusion Therefore, the response surface methodology is feasible for the preparation parameter optimization of phosphorelated modified peanut protein isolate films and the obtained technological conditions have practical application values.
