摘要
目的:建立HPLC法测定人血浆中焦谷氨酸含量的方法。方法:采用HPLC色谱法,色谱柱为Agilent ZORBAX SB-Aq C18柱(4.6 mm×250 mm,5μm),流动相为乙腈-辛烷磺酸钠溶液,梯度洗脱,流速为1.0 m L·min-1,检测波长为210nm,柱温35℃,进样体积20μL。结果:焦谷氨酸在0.0025~0.02 mmol·L-1范围内线性关系良好(R=0.9998),加标回收率在97.2%~111.5%,RSD在0.7%~1.7%。结论:该法简便准确,可用于血浆中焦谷氨酸的测定。
Objective: A HPLC method for the determination of Pyroglutamic Acid in human plasma was established. Methods: Chromatographic separation was performed in Agilent ZORBAX SB-Aq C18 column(4. 6 mm × 250 mm,5 μm) with an eluent consisting of acetonitrile and sodium 1-octanesulfonate solution at a flow rate of 1. 0 ml. min-1. The eluent was monitored at 210 nm and the injection volume was 20 μL. Results: The linear range was 0. 0025 ~ 0. 02 mmol·L-1(r2= 0. 9997),and the recovery range was 97. 2% ~ 111. 5%with RSD 0. 7% ~ 1. 7%. Conclusion: The method was simple,accurate,which can be used for the determination of Pyroglutamic Acid in human plasma.
