摘要
[目的]探讨存活素Survivin、凋亡抑制因子Livin和磷酸酶基因PTEN在结肠正常黏膜组织、腺瘤组织、癌旁组织及癌组织中的表达的变化规律及临床意义。[方法]采用逆转录聚合酶链反应(reverse transcription-PCR,RT-PCR)检测收集到的结肠正常黏膜组织、腺瘤组织、癌旁组织及癌组织各30例中Survivin、Livin和PTEN mRNA的表达水平,分析不同组织中表达水平的差别。[结果]结肠癌组织中Survivin、Livin mRNA表达水平与腺瘤组织比较明显升高,有显著性差异(P<0.05),而PTEN mRNA的表达水平与腺瘤组织比较明显降低,有显著性差异(P<0.05)。Survivin、Livin mRNA水平在正常黏膜组织、腺瘤组织、癌旁组织、癌组织为逐渐增高趋势,而PTEN mRNA为逐渐下降趋势,且Livin在中低分化癌组织中表达高于高分化癌组织,PTEN则在高分化癌组织中表达高于中低分化癌组织。[结论]Survivin、Livin和PTEN在结肠腺瘤到癌的转变过程中起着重要作用。
[Objective]To investigate the expression and the clinical significance of Survivin, Livin and PTEN in normal colon mucous membrane, colon adenoma, colonic carcinoma and their adjacent tissues. [Methods]The reverse transcriptase real-time fluorescent quantitative PCR technique was utilized to detected the mRNA of Survivin, Livin and PTEN in a colonic adenoma group, a colonic carcinoma group, their adjacent tissues, and a normal control group. [Results]The rates of positive expression of Survivin and Livin mRNA in the colon carcinomas were higher than that in colon villous adenoma(P〈0.05). The expressions of Survivin and Livin mRNA in normal colon mucous membrane, the carcinoma adjacent tissues, colon canalicular adenoma and colon carcinomas were gradually increased,the expressions of PTEN mRNA were gradually decreased. The expression of Livin in moderately poorly differentiated cancer tissues was higher than that in well differentiated cancer tissues,while PTEN in high differentiated cancer tissues was higher than that in moderately poorly differentiated cancer tissues. [Conclusion]Survivin,Livin and PTEN play an important role in process carcinomatous change of colon mucous membrane.
