摘要
目的制备灯盏花素维生素E聚乙二醇琥珀酸酯(VE-TPGS)聚合物胶束,并对其进行表征及体外评价。方法采用薄膜溶剂挥发法制备TPGS聚合物胶束,采用粒径测定仪、透射电镜、X-射线衍射对其进行表征;采用紫外分光光度法测定胶束的包封率和载药量;采用动态膜透析法考察载药胶束的体外释药特性。结果薄膜溶剂挥发法制备的胶束呈球形或类似球形,平均粒径为(20±2.62)nm,平均包封率和载药量分别为(90±2.14)%和(5.8±0.19)%;体外释放结果表明,制备的TPGS具有一定的缓释作用。结论该胶束制备工艺简单,其粒径、包封率、载药量均可控,具有缓释作用。
Objective To prepare breviscapine-loaded Vitamin E-TPGS polymeric micelles and characterize and evaluatethemin vitro.Methods The breviscapine-loaded Vitamin E-TPGS polymeric micelles were prepared using the film solvent volatilization method and were characterized by the particle size tester,transmission electron microscope and XRD.The drug loading and entrapment efficiency were determined by UV spectrophotometry.The release characteristics of polymeric micelles in vitro were investigated with a dynamic membrane dialysis method.Results Polymeric micelles was spherical or nearly spherical.The average diameter,entrapment efficiency and drug loading of the polymeric micelles were(20±2.62)nm,(90±2.14)%and(5.8±0.19)%,respectively.The release results showed that the prepared polymeric micelles had some release effect in vitro.Conclusion This preparation process of micelles is simple,and the particle size,encapsulation rate and drug loading are controllable with a sustained release effect.
