红花黄色素A对大鼠骨髓源性内皮祖细胞促血管新生作用研究 被引量:10

摘要 目的探讨红花黄色素A对大鼠骨髓源性内皮祖细胞的促血管新生活性。方法体外分离培养大鼠骨髓源性内皮祖细胞,观察红花黄色素A低、中、高(20、40、60μg·ml^(-1))剂量对骨髓源性内皮祖细胞增殖、黏附、迁移及血管形成能力的影响,以及对骨髓源性内皮祖细胞中促血管新生相关因子蛋白表达的影响。结果与空白对照组比较,不同浓度红花黄色素A可显著促进骨髓源性内皮祖细胞的增殖、黏附、迁移及血管形成能力,骨髓源性内皮祖细胞中促血管新生因子血管内皮生长因子及其受体,碱性成纤维细胞生长因子,血管生成素1的蛋白表达水平也显著上升,且均呈浓度依赖性。结论红花黄色素A具有调控骨髓源性内皮祖细胞促血管新生的作用,该作用可能与其上调血管内皮生长因子及其受体VEGFR-2、碱性成纤维细胞生长因子及血管生成素1的表达密切相关。 Objective To investigate the proangiogenic activities of safflor yellow A(SYA)on bone marrow-derived endothelial progenitor cells(EPCs)in rats.Methods Bone-marrow-derived EPCs were isolated from rats and cultured.EPCs were treated with SYA at different concentrations(20,40 and 60μg·ml-1).The capacity of EPCs’ proliferation,adhesion,migration,tube formation and proangiogenic cytokines expressions were observed in vitro.Results Compared with the control group,different concentrations of SYA could promote the proliferation,adhesion,migration and tube formation of EPCs significantly.Furthermore,the protein expressions of vascular endothelial growth factor(VEGF),vascular endothelial growth factor receptor-2(VEGFR-2),basic fibroblast growth factor(bFGF)and angiopoietin-1(Ang1)were also upregulated markedly in a concentration-dependent manner.Conclusion SYA has potent proangiogenic properties by regulating functions of EPCs.This effect could be attributed to an increased formation of VEGF,VEGFR-2,bFGF and Ang1.
