针刺对组胺致痒小鼠5-羟色胺及其受体表达的影响

Effect of acupuncture on expression of 5-HT and its receptors in mice with histamine-induced pruritus

目的探讨针刺对组胺致痒模型小鼠的瘙痒行为、血清5-羟色胺(5-HT)及延髓5-HT受体表达的影响。方法将30只C57B/6J小鼠随机分为模型组、针刺组和生理盐水组,每组10只。模型组和针刺组均予颈背部皮下注射磷酸组胺进行造模,造模后模型组不做处理,针刺组给予针刺双侧血海、曲池及合谷穴,采用提插和捻转手法刺激,每日1次,针刺2 d;生理盐水组予颈背部皮下注射生理盐水后不做处理。连续观察并记录各组小鼠搔抓次数,采用ELISA及Western blot方法检测5-HT及其受体表达情况。结果模型组和针刺组小鼠造模第1,2,3天搔抓次数均明显多于生理盐水组(P均<0.05);针刺组小鼠造模第1,2天搔抓次数比较差异无统计学意义(P>0.05),但造模第3天搔抓次数明显少于第1,2天及模型组(P均<0.05)。模型组小鼠血清5-HT水平及延髓5-HT受体表达水平均明显高于生理盐水组(P均<0.05),针刺组小鼠血清5-HT水平明显低于模型组(P<0.05),针刺组与模型组小鼠延髓5-HT受体表达水平比较差异无统计学意义(P>0.05)。结论针刺对组胺致痒小鼠的搔抓行为具有抑制作用,下调血清5-HT水平可能是其机制之一。

Objective It is to investigate the effect of acupuncture on pruritus behavior,and the expression of serum 5-hydroxytryptamine（ 5-HT） and medulla oblongata 5-HT receptor in histamine-induced pruritus mice. Methods Thirty cases of C57 B/6 J mice were randomly divided into model group,acupuncture group and saline group,with 10 cases in each group. Both the model group and the acupuncture group were injected with histamine phosphate subcutaneously into the neck and back. After modeling,the model group does not do the treatment. Acupuncture group was given acupuncture at both sides of acupuncture point of Xuehai,Quchi and Hegu,the manipulations of lifting,inserting and twisting were used for stimulation,once a day for 2 days. The normal saline group were received subcutaneous injection of normal saline into the neck and back without other treatment. The scratch times of mice in each group were observed and recorded,and the expression of 5-HT and its receptor was detected by ELISA and Western blot. Results The scratch times in the model group and acupuncture group were significantly higher than those in the normal saline group on the first,second and third days（ all P〈0. 05）. There was no significant difference in scratching times between the first and second days in the acupuncture group（ all P〉0. 05）,however,the scratch times on the third day were significantly less than those on the first,second days and the model group（ all P〈0. 05）. The expression levels of serum 5-HT and medulla oblongata 5-HT receptor were significantly higher in the model group than those in the saline group（ all P〈0. 05）,The serum 5-HT level in acupuncture group was significantly lower than that in the model group（ P〈0. 05）,there was no significant difference in the expression of 5-HT receptor between the acupuncture group and the model group（ P〉0. 05）. Conclusion Acupuncture has inhibitory effect on scratching behavior in mice with histamine-induced pruritus,lowering the level of serum 5-HT may be one of the mechanisms.
