摘要
采用碱性蛋白酶、纤维素酶和碱性蛋白酶复合纤维素酶对新鲜双孢蘑菇子实体进行水解,设计L9(34)正交试验考查温度、pH值、酶添加量和时间对蛋白水解度的影响,制备蛋白水解度最高的酶解液,利用GC-MS对其中的挥发性物质进行分析。结果表明,纤维素酶最佳酶解参数为温度50℃,pH值5.0,酶添加量0.8%,时间120 min;碱性蛋白酶最佳酶解参数为温度40℃,pH值10.5,酶添加量0.9%,时间150 min。用碱性蛋白酶复合纤维素酶水解双孢菇后,水解度达20.80%,共有33种挥发性成分。
The fruit body of Agaricus bisporus was hydrolyzed by enzyme,and the effect of temperature,p H value,enzyme dosage and time on the degree of proteolysis were investigated by L9(34)orthogonal experiment. The volatile components of the hydrolyzate were analyzed by gas chromatography-mass spectrometry(GC-MS). The results showed that the optimum enzymatic hydrolysis parameters of cellulase was the temperature 50 ℃,p H 5.0,the amount of enzyme 0.8%,time 120 min. The best enzymolysis parameters of alkaline protease was the temperature 40 ℃,p H 10.5,the amount of enzyme 0.9% and the time 150 min. Using alkaline protease and cellulose hydrolyze the fruit body of Agaricus bisporus,the degree of hydrolysis was20.80%,33 kinds of volatile components were identified.
