摘要
(1)目的观察痛风舒胶囊对急性痛风性关节炎(AGA)大鼠血清炎症因子及氧化应激的影响,初步探讨其作用机制。(2)方法将32只雄性Wistar大鼠随机分为对照组、模型组、秋水仙碱组、痛风舒胶囊组,每组8只。除对照组外,其余各组采取黄嘌呤(XA)灌胃联合腹腔注射氧嗪酸钾(OAPS),同时各药物组给予相应药物干预,对照组、模型组给予无菌生理盐水灌胃,连续7天,并于第5天向每只大鼠踝关节腔注射尿酸钠溶液建立急性痛风性关节炎模型。分别于踝关节腔注射尿酸钠前后测定其踝关节肿胀程度,检测各组大鼠血清白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)及一氧化氮(NO)的含量,观察大鼠踝关节病理组织学变化。(3)结果实验结束时模型组大鼠踝关节肿胀明显(P <0.01,P <0.05),血清IL-1β、TNF-α、MDA、NO水平较对照组均明显升高(P <0.01),SOD含量明显降低(P <0.01),滑膜表面粗糙,大量炎性细胞浸润,增生明显;痛风舒胶囊组、秋水仙碱组踝关节肿胀度明显低于模型组(P <0.01,P <0.05),其IL-1β、TNF-α、MDA、NO活性显著降低(P <0.01,P <0.05),SOD水平升高明显(P <0.01),滑膜细胞增生、炎细胞浸润情况均有所缓解。(4)结论痛风舒胶囊能够明显缓解AGA大鼠关节症状,其作用机制与降低IL-1β、TNF-α水平,抑制MDA、NO的表达,促进SOD的释放有关。
Objective To observe the effects of Tongfengshu capsula on serum inflammatory factors and oxidative stress in rats with acute gouty arthritis and to explore its preliminary action mechanisms. Methods A total of 32 male wistar rats were randomly divided into control group,model group, the colchicine group,and Tongfengshu capsula group,8 rats in each group. Xanthine combined with intraperitoneal injection of oxazine acid potassium for 7 days and the corresponding drugs were administrated in the meantime.The fifth day,ankle cavity was injected of uric acid sodium to establish AGA rat model.During the experiment,the degree of joint swelling in rats was observed before and after the drug intervention.ELISA was used to detect the contents of IL-1β,TNF-α,SOD,MDA and NO,The pathological changes of ankle joint observed. Results The joint swelling degree of rats in the model control group was significantly increased at each time point ( P 〈 0.01 , P 〈 0.05 ).The contents of IL-1β,TNF-α,MDA and NO were significantly increased ( P 〈 0.01 ) and the level of SOD were significantly reduced ( P 〈 0.01 ).The surface of synovial were rough,a large number of inflammatory cells were infiltrated,and synovial cells was obvious hyperplasia.The joint swelling degree of rats in Tongfengshu Capsula group and the colchicine group were lower than the model control group( P 〈 0.01 , P 〈 0.05 ).The contents of IL-1β,TNF-α,MDA and NO were significantly reduced ( P 〈 0.01 , P 〈 0.05 ),while the contents of SOD were significantly increased ( P 〈 0.01 ),with the symptoms of synovial cells hyperplasia and inflammatory cells infiltration were improvement. Conclusion Tongfengshu capsula has good therapeutic effect in rats with acute gouty arthritis and the mechanism may relate to decrease the levels of IL-1β,TNF-α,MDA and NO and strengthen the expression of SOD.
