摘要
抑郁障碍是一种常见的严重精神疾病,具有高致残率和致死率,给社会和家庭造成严重负担。目前,近一半抑郁症患者无法得到有效的治疗。在临床实践中,可以广泛观察到氯胺酮具有明显缓解抑郁症状的作用,然而对其抗抑郁机理的研究尚不明确。笔者主要着眼于氯胺酮抗抑郁作用机理的相关研究进展,重点关注氯胺酮抗抑郁作用的短暂急性期和持续的延迟期机制的复杂相互作用的生物学基础,根据已有文献和前期研究结果提出可行的预设方案,旨在为氯胺酮抗抑郁作用的临床应用提供科学依据。
Depressive disorder is a common serious mental illness,with significant disability rate and mortality,which has caused serious burden to society and family. At present,nearly half of the depressive patients are unable to get effective treatment. In clinical practice,ketamine was widely observed to alleviate depressive symptoms effectively. However,its antidepressant mechanism is not clear. Focusing on the progresses in antidepressive mechanism of ketamine,this review summarizes the complex interaction mechanism about acute and delayed effect of ketamine,and puts forward the feasible scheme of design based on the existing literatures and previous researches, which aims to provide scientific basis for clinical application of ketamine as an antidepressant.
