摘要
目的观察丹红注射液对大鼠局灶性脑缺血再灌注后脑血管内皮损伤的保护作用。方法采用线栓大脑中动脉willis环缺血45 min后再灌注的方法将60只SD大鼠造模,术后挑选造模成功的36只均分为随机均分为模型组、奈必洛尔组和丹红组(n=12),另12只未手术的SD大鼠设为对照组作为对照。模型组和对照组尾静脉注射0.9%氯化钠注射液,奈必洛尔组尾静脉注射β1受体阻滞剂奈必洛尔,丹红组尾静脉注射丹红注射液,4组均治疗14 d。采眶静脉血检测白细胞介素-10(IL-10)、内皮细胞一氧化氮(NO)、转录因子(NF-κB)、转录激活蛋白-1(AP-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)和内皮素-1(ET-1)含量,取willis环检测内皮细胞NO合酶(eNOs)。结果丹红组静脉血NF-κB、AP-1、ICAM-1、VCAM-1和ET-1均明显降低,IL-10明显升高,与模型组和奈必洛尔组比较差异有统计学意义(P<0.05);内皮细胞e NOs及静脉血NO含量均升高,与模型组比较差异明显(P<0.05)。结论丹红注射液有与β1受体阻滞剂奈必洛尔相同的脑血管内皮损伤保护作用,但其除可提高内皮细胞e NOs及静脉血NO含量而保护损伤内皮外,还可通过提高IL-10表达而发挥抑制巨噬细胞的抗原提呈功能,降低缺血再灌注后氧化应激造成的血管内皮炎症性损伤,从而改善血管内皮功能紊乱现象。
Objective: To study the protective effect of Danhong Injection on cerebral vascular endothelium injury after focal cerebral ischemia-reperfusion in rats. Methods: 60 SD rats were established by reperfusion after occlusion of the willis ring with a thread occlusion of the middle cerebral artery(MCAO) for 60 minutes. All 36 SD rats were randomly divided into three groups: the model group,Nebivolol group and Danhong group(n = 12),and the other 12 non-operation SD rats were the control group. normal saline was injected into caudal vein in the model group and the control group;β1-blocker Nebivolol was injected into the tail vein of Nebivolol group;Danhong Injection was injected into the tail vein of Danhong group. All the rats in the four groups were treated for 14 days. The levels of interleukin-10(IL-10),endothelial nitric oxide(NO),transcription factor NF-κB,AP-1,ICAM-1,VCAM-1 and ET-1 were detected by orbital vein blood;e NOs was detected by willis ring. Results: The levels of NF-κB,AP-1,ICAM-1,VCAM-1 and ET-1 in venous blood of Danhong group were significantly decreased;IL-10 increased significantly,compared with the model group and Nebivolol group(P〈0.05). Endothelial cell e NOs and venous blood NO content increased,compared with the model group(P〈0.05). Conclusion: Danhong Injection has the same protective effect on cerebral vascular endothelium injury as β1-blocker Nebivolol. It not only protects the injured endothelium by increasing the content of e NOs in endothelial cell and NO in venous blood,but also inhibits the antigen presentation function of macrophages by enhancing the expression of IL-10,and reduce the inflammatory injury caused by oxidative stress after ischemia and reperfusion,and improve the vascular endothelial dysfunction.
