虎杖苷对体外培养人翼状胬肉成纤维细胞增殖的影响 被引量:2

摘要 目的探讨虎杖苷对人翼状胬肉成纤维细胞(HPF)增殖的影响,为治疗和预防翼状胬肉复发找寻新依据。方法取人翼状胬肉成纤维细胞体外培养;将不同浓度的虎杖苷作用于成纤维细胞分别达24 h,48 h和72 h后,MTT法检测虎杖苷对HPF增殖的影响,增生细胞核抗原(PCNA)经免疫组织化学染色后检测细胞生长活性,流式细胞技术检查细胞周期及细胞凋亡。结果 MTT法显示10^(-2) mol/L虎杖苷能抑制HPF的增殖(P<0.05),呈时间依赖性,抑制率分别为71.79%、74.60%、75.26%;抑制细胞表达PCNA(P<0.05)。低浓度即10^(-3),10^(-4)mol/L虎杖苷对HPF抗增殖作用不明显。结论 10^(-2) mol/L虎杖苷能够显著抑制HPF的增殖,提示虎杖苷可用于翼状胬肉外科治疗中。 Objective To investigate the effect of polydatin on human pterygium fibroblasts (HPFs) in vitro,so as to search for treatment & prevention of pterygium and its recurrence. Methods HPF was cultivated in vitro. After treatment with 10^-2,10^-3,10^-4mol/L polydatin,MTT assay was used for detecting the effect,and proliferating cell nuclear antigen (PCNA) were detected by immunohistochemical analysis. The cell cycle and apoptosis were measured by flow cytometry. Results The polydatin of 10^-2mol/L had a significant inhibition on HPF proliferation. There were no differences between the cells treated with 10^-3,10^-4mol/L polydatin and the controls in the MTT assay and flow cytometry (P〉0.05). Conclusion Polydatin at 10^-2mol/L can significantly inhibit the proliferation of HPFs at nontoxic concentration by promoting apoptosis. Therefore,polydatin might be used as a safe choice for pterygium surgery to prevent recurrence.
