摘要
目的探讨膝骨关节炎模型大鼠在穴位敏化的不同时间节点相关穴区局部肥大细胞(mast cells,MC)变化与穴位敏化的关系。方法选取150~160 g的SD雄性大鼠随机分为正常组(N)、生理盐水组(NS)、模型组(KOA)三组,每组按照0天、7天、14天、21天、28天随机再分为五组。在右侧膝关节腔内注射单碘乙酸盐(MIA)制备模型。造模成功后分别于造模的第0天、7天、14天、21天、28天时取1.5 mm×1.5 mm×1.5 mm体积的鹤顶、阳陵泉、委中穴区局部皮肤组织,采用甲苯胺蓝染色法检测MC。结果在造模后的第7天,KOA组大鼠鹤顶穴区MC的数量及脱颗粒率高于N和NS组(P<0.05),MC数量高于0天、21天(P<0.05);第7天、14天MC脱颗粒率均高于0天(P<0.05);造模后第14天,KOA组大鼠阳陵泉穴区MC数量及脱颗粒率明显高于N和NS组(P<0.05),MC数量和脱颗粒率高于同组的第0天和7天(P<0.05);而委中穴MC数量及脱颗粒率则没有明显变化。结论膝骨关节炎模型大鼠相关穴位在疾病过程中某一时间点发生了敏化,敏化效应持续一段时间;穴位敏化是动态变化的过程,并且敏化伴随了穴区MC的募集和脱颗粒;穴位敏化具有时间和空间的特异性。
Objective To investigate the relationship between the changes of local mast cells (MC) and acupoint sensitization in the acupoint sensitization of different time nodes in knee osteoarthritis model rats. Methods Male rats of 150~160 g were randomly divided into normal group (N), normal saline group (NS) and model group (KOA). Each group was randomly divided into five groups according to 0, 7 th , 14 th , 21 st and 28 th day. The model was prepared by intraluminal injection of monoiodoacetate (MIA) into the right knee joint. After the successful modeling, the local skin tissues with the volume of 1.5 mm× 1.5 mm×1.5 mm of Heding (EX-LE2), Yanglingquan (GB34) and Weizhong (BL40) were taken at 0, 7 th , 14 th , 21 st and 28 th day of the modeling. The toluidine blue staining was used to detect MC. Results On the 7 th day after modeling, the number and degranulation rate of MC in Heding acupoints of KOA group were higher than those of N and NS groups ( P 〈0.05), and the number of MC was higher than 0 and 21 st day ( P 〈0.05). The degranulation rate of MC at 7 th and 14 th was higher than 0 day ( P 〈0.05). On the 14 th day after modeling, the number of MC and degranulation rate in Yanglingquan acupoint of KOA group were significantly higher than those in N and NS group ( P 〈0.05). The number of MC and the rate of degranulation were higher than those of the same group on the 0 and 7 th day ( P 〈0.05); while the number of MC and the rate of degranulation in the center were not significantly changed. Conclusion The related acupoints in the knee osteoarthritis model rats were sensitized at a certain point in the disease process, and the sensitization effect continued for a while. The acupoint sensitization was a dynamic change process, and the sensitization was accompanied by the acupoint MC recruitment and degranulation. Acupoint sensitization has time and space specificity.
