摘要
目的分析实时无标记细胞分析系统与台盼蓝染色法分析双酚A(BPA)对小鼠睾丸间质细胞毒性作用,以确定最佳实验方案。方法采用实时无标记细胞分析系统及台盼蓝染色法测定BPA对小鼠睾丸间质细胞的细胞毒性,计算其IC_(50)。结果实时无标记细胞分析法及台盼蓝染色法结果表明,不同浓度BPA使小鼠睾丸间质细胞的活力下降,1 000μmol/L时下降最为明显,IC_(50)分别为704、745μmol/L。但实时无标记系统能够实时监测细胞生长的动态生物学反应。结论实时无标记分析法能够更直观、准确地观察细胞生长情况,弥补台盼蓝染色法的缺点,是一种新的研究细胞毒性的实验方法。
Objective To determine the best experimental scheme by using the real-time cell analysis and Trypan blue staining method in the analysis of the toxicity of bisphenol A on leydig cells in mice. Methods The cyto-toxic effect and IC50 of BPA on mouse leydig cells were measured by real-time cell analysis and Trypan blue staining method. Results The results of real-time cell analysis and Trypan blue staining showed that BPA of different concen-trations reduced the activity of leydig cells in mice,and the most obvious decrease was at 1 000 μmoL/L;the IC50 values were 704 μmol/L and 745 μmol/L, respectively. Real-time cell analysis can monitor the dynamic biological response of cell growth in real time. Conclusion The real-time cell analysis can be more intuitive and accurate in observing the cell growth,and make up for the shortcomings of Trypan blue staining method, which becomes a new experimental method to study cytotoxicity.
