雅连对2型糖尿病大鼠胰岛素抵抗和TNF-α、IL-6、FFA表达的影响

Effects of Ya Lian on Insulin Resistance and Expression of TNF-α,IL-6 and FFA in Type 2 Diabetic Rats

目的:探讨雅连浸渍剂对链脲佐菌素(Streptozotocin,STZ)诱导的2型糖尿病(Type 2 Diabetes Mellitus,T2DM)大鼠血糖、胰岛素抵抗(Insulin Resistance,IR)和肿瘤坏死因子(Tumor Necrosis Factorα,TNF-α)、白介素-6 (Interleukin-6,IL-6)、游离脂肪酸(Free Fatty Acid,FFA)的影响。方法:采用高脂高糖饲料联合浓度1%的STZ 40mg/kg剂量诱导建立T2DM大鼠模型,将60只大鼠随机分为空白组、模型组、西格列汀组、雅连组。观察大鼠体质量,检测空腹血糖,血清胰岛素,血清TNF-α、IL-6、FFA,计算胰岛素抵抗指数。结果:与空白组相比,模型组大鼠体质量减少,空腹血糖明显升高,血清TNF-α、IL-6、FFA水平显著升高,胰岛素抵抗指数明显增高;西格列汀组、雅连组给药3 w后,上述指标明显下降,并且雅连组对空腹血糖,体质量的控制比西格列汀组更稳定,对胰岛素抵抗的改善、降低FFA水平更加明显。结论:雅连可能通过降低炎症因子水平,减轻炎症反应,改善胰岛素抵抗等途径缓解T2DM。

Objective ： To investigate the effects of ya lian dipping agent on blood glucose, insulin resistance （ IR ）, tumor necrosis factor a （TNF-a）, interleukin-6 （IL-6） and free fatty acid （FFAs） induced by streptozotocin （STZ） in rats wdth type 2 diabetes mellitus （T2DM）. Methods： The rat model of T2DM was established by high fat and sugar diet combined with 1% STZ 40mg ？ kg-1. 60 rats wee randomly divided into normal group,model group, sitagliptin group and ya lian group. The body weight,fasting blood glucose, serum insulin, serum TNF-a, IL-6, FFA were measured and insulin resistance index was calculated. Results： Compared ■with normal group,the weight loss and fasting blood glucose in model group increased significantly, the serum TNF- a , IL-6 and FFA increased significantly, and the insulin resistance index increased significantly in the model group. The above indexes decreased signifi-cantly in sitagliptin group and ya lian group, and the control of fasting blood glucose and body weight in ya lian group was more stable than that of sitagliptin group, and the improvement of insulin resistance and the decrease of FFA level were more obvious. Conclusion： The treatment of T2DM by ya lian may be achieved via reducing the level of inflammatory cytokines and the inflammatory reaction and improving insulin resistance.
