养血祛风方对抽动障碍大鼠纹状体GAP-43表达影响的研究 被引量:2

摘要 目的:观察养血祛风方对抽动障碍(Tic Disorders,TD)大鼠行为学特征与纹状体GAP-43和多巴胺受体D1/D2 (DRD1/D2) mRNA表达的影响,探讨其对TD大鼠纹状体突触可塑性影响。方法:采用亚氨基二丙腈(IDPN)复制TD大鼠模型,按随机数字表法将TD模型大鼠分为模型组、西药组及低、中、高剂量中药组,每组各8只,另取8只健康SD大鼠做空白组;给药组TD大鼠予以相应药物灌胃,空白组及模型组则予以等量生理盐水。从刻板运动方面对各组大鼠行为学特征进行评估和比较;持续灌胃8周后,取大鼠纹状体,采用免疫组化SP法检测GAP-43表达水平。结果:刻板运动测试显示大鼠模型能良好模拟TD的特征性行为变化,中药组及西药组刻板运动评分均不同程度低于模型组(P <0. 05)。各组GAP-43表达水平比较,差异具有显著统计学意义(P <0. 001)。其中,模型组GAP-43表达水平明显低于空白组及各给药组(P <0. 05);而各剂量中药组GAP-43表达水平均高于西药组(均P <0. 05);不同剂量中药组之间比较,高剂量组和中剂量组GAP-43表达水平均较低剂量组明显增高(P <0. 05)。结论:养血祛风方可通过上调GAP-43表达水平,在TD大鼠纹状体突触重塑方面发挥正向调节作用。 Objective: To observe the effect of Nourishing Blood and expelling Wind recipe on the behavioral characteristics and the expression of GAP43, DRD1/D2 mRNA in striatum of rats with tic disorder (TD) , and to investigate its effect on synaptic plasticity in striatum of rats with TD. Methods : The rat model of TD was duplicated by intraperitoneal injection of iminodipropionitrile (IDPN),and TD rats successfully modeled were randomly divided into model group, western medicine group, low-, medium-, and high-dose Chinese medicine groups according to the random number table method, with 8 rats in each group and 8 healthy SD rats were used as blank group. TD rats in the administration group were given the corresponding drugs by gavage; the blank group and the model group were given the same amount of nor^nal saline. Assess and compare the behavioral characteristics of rats in the stereotyped move-ment. After 8 weeks of continuous gavage, rat striatum was taken and the expression of GAP^3 was detected by immunohistochemical SP method, and the expression of DRD1/D2 mRNA was detected by RT-PCR. Results: The stereotyped exercise test showed that the rat model could well simulate the characteristic behavior changes of TD, and the scores of stereotyping in the Chinese medicine group and the western medicine group were all lower than the model group ( P 〈 0. 0 5 ) . Compared with the expression level of GAP43 in each group, the difference was statistically significant ( P 〈0. 001) . Among them, the expression level of GAP43 in the model group was significantly lower than that in the blank group and each administration group ( P 〈0. 05) . The expression level of GAP43 in each dose group was higher than that in the western medicine group (all P 〈 0. 05) ; comparison between different doses of TCM groups, high-dose group and middle-dose group GAP43 expression levels in the lower-dose group was significantly higher ( P 〈 0. 05). Conclusion: Nourishing Blood and expelling Wind recipe can positively regulate the synaptic remodeling by increasing the ex-pression of GAP^3 in striatum Effect.
