祖师麻肠溶微丸对结扎坐骨神经所致痛觉敏感大鼠背根神经节P2X4受体及P38MAPK表达的影响 被引量:3

摘要 目的:观察灌胃给予祖师麻肠溶微丸对CCI大鼠的MWT以及背根神经节中P2X4和P38MAPK受体的影响,探究祖师麻肠溶微丸治疗坐骨神经痛的机制。方法:术后第8天按MWT值对位随机分配法分为7组:Control组、Sham组、Model组、Positive Drug组、HEPZ组、MEPZ组和LEPZ组,于术后第8天开始灌胃给药。给药14 d后灌注固定,取背根神经节,免疫组化观察P2X4和P38MAPK的表达情况。结果:Model组在第8、14和20天MWT明显降低(P〈0.01)。与Model组相比,其余各组第14天MWT值出现显著差异(P〈0.01),20 d以后差异持续存在(P〈0.01)。免疫组化P2X4在背根神经节中的表达情况,各组均与Model组有统计学差异(P〈0.01),P38MAPK的表达情况除Positive Drug组外,各组与Model组相比,有明显差异(P〈0.05)。结论:口服祖师麻肠溶微丸可明显改善CCI模型,提高MWT,且背根神经节中P2X4和P38MAPK受体表达下调,提示祖师麻肠溶微丸有治疗坐骨神经痛的作用,其止痛的效果与P2X4-P38MAPK通路有关。 Objective:To observe of Zushima Enteric-coated Pellets on CCI rats MWT value and effect of P2 X4 and P38 MAPK receptors in the dorsal root ganglion,to explore the mechanism of Zushima Entericcoated Pellets in the treatment of sciatica. Methods:Eighth days after operation,the rats were divided into 7 groups according to MWT value with contraposition random distribution method,control group,sham group,model group,positive drug group,HEPZ group,MEPZ group and LEPZ group,which were given drugs administration eighth days after the operation. Perfusion 14 days later,take the dorsal root ganglion,immunohistochemistry was used to observe the expression of P2 X4 and P38 MAPK receptors. Results:MWT value decreased significantly at 8,14 and 20 days in model group(P〈0.01). Compared with model group,the MWT values of the other groups showed significant difference on the fourteenth day(P〈0.01). After 20 days,the difference persisted(P〈0.01). Expression of IHC P2 X4 in dorsal root ganglion,each group has significant difference with model group(P〈0.01). The expression of P38 MAPK except positive drug group,each group had significant difference with model group(P〈0.05). Conclusion:Taking Zushima Entericcoated Pellets can obviously improve CCI model,improve the MWT value,and makes the down-regulation of P2 X4 and P38 MAPK receptor expression in the dorsal root ganglion,witch suggested that ZushimaEnteric-coated Pellets has therapeutic effect on sciatica that the analgesic effect is related to the P2 X4-P38 MAPK pathway.
