糖肾康浓缩丸对糖尿病肾病大鼠保护作用及机制研究

Study on the Protective Effect and Mechanism of Tangshenkang Concentrated Pill on Diabetic Nephropathy Rats

目的：探讨糖肾康浓缩丸对糖尿病肾病模型大鼠的保护作用及作用机制。方法：将72只大鼠随机分为假手术组、贝那普利组、模型组、糖肾康高剂量组、糖肾康中剂量组、糖肾康低剂量组。采用肾单侧切除加高糖高脂饮食联合STZ诱导糖尿病肾病大鼠模型,成模后假手术组及模型组用生理盐水灌胃,治疗组分别以糖肾康浓缩丸和盐酸贝那普利灌胃,4周取血、留尿、取肾组织,检测大鼠血糖、总胆固醇、甘油三酯、尿微量白蛋白、血肌酐、尿素氮、相对肾重及肾组织超氧化物歧化酶、丙二醛、肿瘤坏死因子-α,光镜观察肾脏病理学变化,比较各组指标差异。结果：各治疗组较模型组的血糖、血脂、肾功能、病理学变化以及氧化应激和炎症指标均有不同程度的改善,其中糖肾康中剂量效果改善最显著,明显优于贝那普利组,差异具有显著性（P〈0.05）。结论：糖肾康浓缩丸通过抑制炎症和氧化应激反应延缓糖尿病肾病进程。

Objective：To investigate the protective effect and mechanism of Tangshenkang Concentrated Pill on diabetic nephropathy rats. Methods：72 rats were randomly divided into sham operation group,benazepril group,model group,high dose,medium and low doses of Tangshenkang. The model of diabetic nephropathy rats was induced by unilateral nephrectomy and hyperlipidemia and high glucose diet. The sham operation group and model group were perfused with saline,while the treatment groups were perfused with Tangshenkang Concentrated Pill and Benazepril. After 4 weeks of treatment,blood glucose,total cholesterol and triglyceride,urinary albumin,serum creatinine,urea nitrogen,the relative renal weight and renal tissue superoxide dismutase,malondialdehyde and tumor necrosis factor-alpha were measured,and pathological changes of kidney were observed by light microscopy,and the differences of the indexes of every group were compared. Results：Compared with the model group,the blood glucose,blood lipid,renal function,pathological changes,oxidative stress and inflammatory indexes of the treatment groups were improved in different degrees,among them,the curative effect of the middle dose group was the most significant,there was a significant difference between the benazepril group and middle dose group（P〈0.05）.Conclusion：Tangshenkang Concentrated Pill can delay the progress of diabetic nephropathy by inhibiting inflammation and oxidative stress.
