不同剂量艾司洛尔对脓毒症心肌病大鼠TNF-α和IL-10的影响

Effects of different does of esmolol on TNF-α and IL-10 expression in septic cardiomyopathy rats

目的探讨艾司洛尔在脓毒症心肌病大鼠心脏功能保护中的作用。方法选取3个月的雄性大鼠50只,随机分为:空白对照组(K组)、模型组(L组)、干预组(T组)(又分T1、T2、T3 3个亚组),各10只,观察药物干预后3、6和12h各组大鼠血浆中肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)的水平变化。结果药物干预后3、6、12h,L组大鼠血浆中TNF-α和IL-10水平明显高于K组,差异有统计学意义(P<0.05)。干预组中3个亚组,随着艾司洛尔药物剂量增大,血浆TNF-α水平下降越明显,IL-10水平变化与艾司洛尔剂量变化无关。L组与T组3个亚组比较,组间TNF-α水平差异均有统计学意义(P<0.05),而各组间IL-10水平差异无统计学意义(P>0.05)。结论脓毒症时促炎和抑炎反应同时被激发,艾司洛尔能减少脓毒症心肌病大鼠血浆中的促炎因子的产生,同时能减慢心率,降低心肌细胞氧耗,保护心脏功能,改善预后。

Objective To explore the effect of esmolol on cardiac function protection in septic cardiomyopathy rats.Methods There were 50 male rats of 3 month in age were selected,and randomly divided into the following groups：blank control group（K group）,model group（L group）,intervention group（T group）（divide into three subgroups T1,T2,T3）,and 10 rats in each group.At 3 h,6 hand 12 hafter drug intervention,the levels of TNF-α and IL-10 were detected.Results At 3 h,6 hand 12 hafter drug intervention,the levels of TNF-α and IL-10 were significantly higher in group L than group K,the differences were statistically significant（P 0.05）.Among three subgroups in the intervention group,with the dose of esmolol increased,the level of TNF-α dropped significantly,but the level of IL-10 was not related to the dose of esmolol.Among group L and the three subgroups in group T,the differences in TNF-level between groups were statistically significant（P 0.05）,but the differences in IL-10-level between groups were not statistically significant（P 0.05）.Conclusion Proinflammatory and anti-inflammatory responses to sepsis are stimulated simultaneously.Esmolol can reduce the production of proinflammatory factors in the plasma of septic cardiomyopathy rats,decrease heart rate,reduce oxygen consumption of cardiac myocytes,protect heart function and improve prognosis.
